摘要
目的:探讨沉默信息调节因子3(Sirt3)及过氧化物酶体增殖物激活受体γ辅助活化因子1α(PGC-1α)在子宫内膜样腺癌组织中的表达、临床意义及两者的相关性。方法:采用免疫组织化学法检测子宫内膜样腺癌组织59例、子宫内膜增生症10例、正常子宫内膜组织10例中的Sirt3和PGC-1α的表达情况,分析其与临床病理参数的关系。结果:子宫内膜样腺癌组织中Sirt3的阳性表达率(67.8%)和PGC-1α的阳性表达率(54.2%)显著高于正常子宫内膜组织及子宫内膜增生症,差异有统计学意义(P<0.05)。在子宫内膜样腺癌组织中,Sirt3蛋白的阳性表达率在III+IV期组高于I+II期组、肌层浸润深度≥1/2组高于<1/2组,差异有统计学意义(P<0.05);PGC-1α蛋白的阳性表达率在III+IV期组高于I+II期组、肌层浸润深度≥1/2组高于<1/2组、有远处转移组高于无远处转移组,差异有统计学意义(P<0.05);在子宫内膜样腺癌组织中Sirt3和PGC-1α表达呈正相关(rs=0.313,P<0.05)。结论:Sirt3与PGC-1α在子宫内膜样腺癌组织中呈高表达,其表达水平越高,子宫内膜样腺癌恶性程度越高。Sirt3与PGC-1α的表达呈正相关,两者的协同作用在子宫内膜样腺癌的发生与发展中起到重要作用。
Objective: To investigate the expression and clinical significance of Sirt3 and PGC-1α in endometrial adenocarcinoma. Methods: A total of the data of 59 paraffin-embedded specimens of endometrial adenocarcinoma tissues,10 paraffin-embedded specimens of proliferative endometrium tissues,10 paraffin-embedded specimens of normal endometrium tissues were collected. Immunohistochemistry was used to detect the expression of Sirt3 and PGC-1α. Results: In endometrial adenocarcinoma tissues,Sirt3 positive expression rate( 67. 8%),and PGC-1α positive expression rate( 54. 2%) were significantly higher than that of normal endometrium and hyperplasia endometrium tissue,and the difference was statistically significant( P〈 0. 05). In endometrial adenocarcinoma tissues,Sirt3 protein positive expression rate of III + IV stage group was higher than I + II stage group,and the muscle layer infiltration depth≥1/2 was higher than 〈1/2 group,and the difference was statistically significant( P〈 0. 05). PGC-1α protein positive expression rate of III + IV stage group was higher than I + II stage group,and the muscle layer infiltration depth≥1/2 was higher than 〈1/2 group,and distant metastasis group was higher than that of no metastasis,and the difference was statistically significant( P 〈0. 05). There was positive correlation between Sirt3 and PGC-1α in endometrial adenocarcinoma tissues( rs= 0. 313,P 〈0. 05). Conclusion: Sirt3 and PGC-1α are highly expressed in endometrial adenocarcinoma,and the expression level is higher,the malignant degree of endometrial adenocarcinoma is higher. There is positive correlation between Sirt3 and PGC-1α,and the synergistic effect plays an important role in the occurrence and development of endometrial adenocarcinoma.
