摘要
利用味精合成了味精类脂,构建了可输送紫杉醇的脂质体。谷氨酸单钠盐(味精的核心组分)的氨基进行Boc基团保护后,将两个羧基通过EDC缩合法分别与油胺反应形成酰胺,最后用三氟乙酸溶液脱除Boc保护基团,获得目标产物——味精类脂,总收率为47.0%。在有/无紫杉醇条件下,味精类脂都可通过水浴超声法组装制备脂质体。激光粒度分析仪检测结果表明,紫杉醇脂质体的平均粒径为134.8±4.9 nm(小于空白脂质体的平均粒径157.0±16.5nm),平均表面电位为-5.5±0.3 mV(低于空白脂质体的表面电位+88.1mV)。透射电子显微镜下,紫杉醇脂质体与空白脂质体均为球形结构。细胞实验结果证实:味精类脂构建的脂质体可以用作药物输送载体。
A lipoid was synthesized with the monosodium glutamate( MSG)and used to construct liposomes as carrier for taxol deliv-ery. First, the amino group of MSG (the raw material) was protected by the Boc group. Then, both of its two carboxyl groups were modified with oleylamine molecules by the method of 1-ethyl-( 3-dimethylaminopropyl) carbodiimide hydrochloride( EDC) condensa-tion. At last,the Boc group of the modified glutamic acid was removed by TFA solution,and the lipoid based on monosodium gluta- mate( target product) was obtained. The final yield was 47. 0%. The liposomes could be constructed by the ultrasonic bath method, used the glutamic lipoids alone or together with taxol. The average size of taxol liposomes was 134. 8 ±4. 9 n m,and was smaller than the blank liposome(157. 0±16. 5nm),detected with a laser particle size analyzer. The average surface potential voltage of the taxol liposomes was-5. 5 ±0. 3 mV and the blank liposomes was 88. 1 ±0. 6mV. The shapes of taxol liposomes and blank liposomes both were spherical scanned by the transmission electron microscopy. Our results confirmed that the liposomes constructed with glutamic lipoids could be used as the medicine delivery system.
