Determination of Contents of Four Components in Lanqin Oral Liquid by HPLC Wavelength Switching Method

摘要 目的建立HPLC波长切换法同时测定蓝芩口服液中(R,S)-告依春、黄芩苷、栀子苷、盐酸小檗碱4种成分的含量。方法采用Agilent ZORBAX Eclipse XDB-C18(4.6mm×50mm,3.5μm)色谱柱,以乙腈(A)-磷酸盐缓冲液(B)(磷酸氢二钠2.3996g溶解于1000ml水中,用磷酸调pH值至3.0)为流动相,梯度洗脱(0~3min,2%A;3~13min,2%~20%A;13~20min,20%~45%A);波长:0~5min,245nm;5~12min,238nm;12~20min,265nm;流速:1.0ml·min^(-1);柱温:30℃;进样量:10μl。结果(R,S)-告依春、栀子苷、黄芩苷、盐酸小檗碱的线性范围分别是0.005 780~0.1156μg(r=0.9997)、0.2251~4.502μg(r=0.9999)、0.1070~2.139μg(r=0.9994)、0.027 29~0.5458μg(r=0.9994);平均加样回收率(n=6)分别为98.53%、97.79%、97.05%、98.78%,RSD分别为1.31%、1.13%、1.23%、0.67%。结论该方法操作简单,快速,重复性好,可用于蓝芩口服液的质量控制。 Objective To develop an HPLC method for simultaneous determination of four components,including(R,S)-goitrin,geniposide,baicalin and berberine hydrochoride,in Lanqin oral liquid.Methods The Agilent ZORBAX Eclipse XDB-C18(4.6mm×50mm,3.5μm)column was used.The mobile phase was acetonitrile(A)-phosphate buffer solution(B)(containing 2.3996 g disodium hydrogenphosphate in 1000 ml water and pH adjusted to 3.0 with phosphoric acid)with gradient elution(0-3min,2%A;3-13 min,2%-20% A;13-20 min,20%-45% A)at a flow rate of 1.0ml·min^-1.The detection wavelengths were 245 nm in 0-5min,238 nm in 5-12 min,and 265 nm in 12-20 min respectively.The column temperature was 30℃.Results The linear ranges of(R,S)-goitrin,geniposide,baicalin and berberine hydrochoride were 0.005 780-0.1156μg(r=0.9997),0.2251-4.502μg(r=0.9999),0.1070-2.139μg(r=0.9994),and 0.027 29-0.5458μg(r=0.9994)respectively,the average recoveries(n=6)were 98.53%,97.79%,97.05% and 98.78%,respectively,and the RSD was 1.31%,1.13%,1.23% and0.67%,respectively.Conclusion This method is easy,quick,accurate and reproducible,which can be used for quality control of Lanqinoral liquid.
