摘要
目的:比较水合氯醛及水合氯醛和乌拉坦的1:1混合液作为麻醉剂对Wistar大鼠血管性痴呆(VD)模型复制的影响。方法:分别用水合氯醛及不同剂量的水合氯醛和乌拉坦1:1混合液对实验用Wistar大鼠进行麻醉后造模,模型复制采用双侧颈总动脉缺血再灌注后结扎剪断的方法。比较其麻醉起效时间、持续时间、麻醉死亡率,并于造模后3天使用Morris水迷宫对模型大鼠进行鉴定,比较各模型组之间的行为学差异。结果:在实验过程中,混合液0.5mL/100g组大鼠麻醉起效时间快于10%水合氯醛0.4mL/100g组(P<0.05),且麻醉持续时间较单用水合氯醛组长(P<0.05);Morris水迷宫模型鉴定结果,混合液0.5mL/100g组模型大鼠逃避潜伏期比单用水合氯醛组长(P<0.05);3组间麻醉死亡率对比无明显差异。结论:水合氯醛和乌拉坦的1:1混合液以0.5mL/100g的剂量使用,麻醉起效快、持续时间长、麻醉死亡率低,且大鼠VD模型复制的结果优于单用水合氯醛麻醉组,适用于短期研究的Wistar大鼠的VD模型复制。
Objective: To compare the anesthetic effectiveness of chloral hydrate, urethane and Chloral hydrate 1:1 mixture on making Wistar rats vascular dementia model. Method: using different dose of anesthetic on Wistar rats and then making models. The model rats were made by clipping and cutting off bilateral carotid artery permanently after blocking it three times repeatedly. Comparing the anesthetic onset times pan, duration, mortality and then checking the model rats three days after operation by using Morris water maze to analyze the difference of praxeology between dif- ferent models. Results: The group of 0.5mL/100gof mixture of chloral hydrate and urethane anesthetized rats had faster ouser time and longer duration than single chloral hydrate group (P〈0.05), Morris water maze showed that the group of 0.SmL/100g of mixture of chloral hydrate and urethane anesthetized rats had longer latency than single chloral hydrate group (P〈0.05), but the group of 0.SmL/100g of mixed anesthetic has higher death rate than the other two groups. Con- clusion: 1:1 mixture of chloral hydrate and urethane by using the dose of 0.SmL/100g shows outstanding anesthetic ef- fect in the initiation and better VD rats' model making effect and was more desirable for making short-term VD rats'model.
