摘要
目的研究转移黏附基因(MTDH)在乳腺癌组织中的表达及与肺转移的相关性,为进一步研究乳腺癌侵袭转移奠定基础。方法将40只裸小鼠随机分为空白对照组及乳腺癌模型组a、b、c,分别于接种第10、15、22日处死模型组a、b、c和空白对照组(与模型组c同时处死),取原位瘤及肺组织,肺表面结节计数,免疫组化检测瘤组织中MTDH表达水平。结果模型组a、b、c乳腺癌组织中MTDH阳性表达率分别为30.3%、62.7%、88.2%,表达逐渐增高;对3组裸鼠肺转移数目进行统计发现,有肺转移者,MTDH表达量更高(χ~2=7.473,P<0.01),且肺转移数目越多,其表达量越高(Spearman=0.859,P<0.01)。结论 MTDH在乳腺癌组织中的表达显著高于正常乳腺组织,且与肿瘤的进展、肺转移的数目呈正相关,提示MTDH与乳腺癌的发生、发展密切相关,与乳腺癌肺转移密切相关。
Objective To investigate the expression of metadherin (MTDH) in breast cancer tissue and its correlation with pulmonary metastasis, and lay the foundation for further study on mechanism of invasion and metastasis of breast cancer. Methods Totally 40 nude mice were randomized into a blank control group and 3 breast cancer model group a, b, and c. Mice in model group a, b, c and the blank control group (with group c at the same time) were respectively sacrificed on 10th day, 15th day, and 22th day after modeling. Tumor tissue in situ and lung tissue were collected to detect the expression level of MTDH, and count the lung nodules. Results The positive expression rate of MTDH in the breast cancer tissue of model group a, b, and c was 30.3%, 62.7%, and 88.2%, increasing gradually; Statisticed analysis of the number of lung metastasis in the three groups of nude mice found that higher expression of MTDH in the pulmonary metastasis group (x2 = 7.473, P 〈 0.01). The more the pulmonary metastasis, the higher the expression quantity (Spearman = 0.859, P 〈 0.01). Conclusion Expression of MTDH in breast cancer tissue is significantly higher than in normal breast tissue, and is positively correlated with the progress of tumor and the number of lung metastasis, suggesting MTDH is closely related to the occurrence and development of breast cancer, and associated with the pulmonary metastasis of breast cancer.
