超滤法结合高效液相色谱法测定藤黄酸在不同种属血浆中的血浆蛋白结合率 被引量:1

摘要 目的考察藤黄酸在小鼠、大鼠、家兔和健康人血浆中的血浆蛋白结合率。方法采用超滤法结合高效液相色谱法对藤黄酸与小鼠、大鼠、家兔和健康人血浆的蛋白结合率进行测定。结果藤黄酸在2.65、5.30和10.60 mg·L^(-1)浓度水平与小鼠的蛋白结合率分别为(34.72±2.58)%、(34.09±1.20)%和(33.96±1.25)%,与大鼠的蛋白结合率分别为(34.99±2.35)%、(35.42±2.18)%和(34.75±0.78)%,与家兔的蛋白结合率分别为(33.53±2.20)%、(35.35±1.01)%和(33.83±1.15)%,与健康人血浆的蛋白结合率分别为(34.42±2.28)%、(34.23±1.62)%和(34.62±2.70)%。结论藤黄酸在本实验研究浓度范围内与小鼠、大鼠、家兔和健康人血浆均属于低强度结合,不同种属血浆中各浓度的藤黄酸血浆蛋白结合率无显著差异,且藤黄酸蛋白结合率与药物浓度无明显依赖关系。 Objective To determine the plasma protein binding rate of gambogic acid in the plasma of mouse, rat, rabbit and human. Methods Ultraflitration and HPLC were employed to determine the plasma protein binding rate of gambogic acid. Results The plasma protein binding rates of gambogic acid in the mouse plasma at of 2.65, 5.30 and 10.60 mg L-1 were (34.72±2.58)%, (34.09± 1.20)% and (33.96± 1.25)%; in the rat plasma were (34.99 ± 2.35)%, (35.42 ± 2.18)% and (34.75 ± 0.78)%; in the rabbit plasma were (33.53 ± 2.20)%, (35.35 ±1.01)% and (33.83± 1.15)%; while in the human plasma were (34.42 ±2.28)%, (34.23 ± 1.62)% and (34.62 ±2.70)%, respectively. Conclusion Plasma protein binding characteristics of gambogic acid show low intensity binding in the plasma of mouse, rat, rabbit and human.
