摘要
目的观察参知健脑胶囊单次灌胃对小鼠中枢神经系统功能的影响。方法参知健脑胶囊单次灌胃给予小鼠,共4项试验,分别是自主活动、体温和抓力试验以及戊巴比妥钠阈下催眠试验。其中,自主活动、体温和抓力试验共用40只小鼠,采用区组随机方法分为参知健脑胶囊低剂量组、中剂量组和高剂量组(60、180、540 mg/kg)和阴性对照组(灭菌注射用水),每组各10只,雌雄各半。灌胃给药,于给药后(90±5)min、24 h±5 min进行一般行为观察及自主活动试验;给药后(90±15)min、24 h±15 min进行抓力测试;给药后(90±20)min、24 h±20 min进行体温测试。戊巴比妥钠阈下催眠试验共用50只小鼠,采用区组随机方法分为参知健脑胶囊低剂量组、中剂量组和高剂量组(60、180、540 mg/kg)、阴性对照组(灌胃给予灭菌注射用水)和阳性对照组(腹腔注射地西泮注射液),每组各10只,雌雄各半。阳性对照组于给药后(30±5)min腹腔注射戊巴比妥钠,其余各组于给药后(90±5)min腹腔注射戊巴比妥钠,均测试给药后30 min内小鼠翻正反射消失数。结果给药后的90 min内和24 h内观察,动物一般行为、姿势、步态、流涎、肌颤、瞳孔未见异常。参知健脑胶囊各剂量组小鼠的自主活动数、小鼠抓力、体温和翻正反射消失数与阴性对照组比较,差异无统计学意义(P>0.05)。戊巴比妥钠阈下催眠试验中阴性对照组、低剂量组、中剂量组和高剂量组动物均未出现翻正反射消失,阳性对照组10只动物翻正反射全部消失。结论在本试验条件下,参知健脑胶囊单次灌胃给予小鼠,在60、180、540 mg/kg剂量范围内对小鼠中枢神经系统功能无明显影响,与戊巴比妥钠无协同作用。
Objective To observe the effects of Shenzhijiannao Capsules on the central nervous system in mice after single-pass administration. Methods There were four tests after single-pass administration, which respectively were autonomic activity, body temperature, the holding power, and nembutal subthreshold hypnotic tests. 40 mice were shared in autonomic activity, body temperature and the holding power tests, and according to block random method they were divided into four groups by low-dose group, middle-dose group and high-dose group of Shenzhijiannao Capsules (60, 180, 540 mg/kg) and negative control group (sterile water for injection), with 10 animals in each group, all half male and female. The general behavior observation and autonomic activity test were executed (90±5)min, 24 h±5 min af- ter intragastric administration, the holding power test were executed (90±15) min, 24 h±15 min after intragastric admin- istration, and body temperature test (90±20) min, 24 h±20 min. 50 mice were observed in nembutal subthreshold hyp-notic test, and according to block random method they were divided into five groups by low-dose group, mid- dle-dose group and high-dose group of Shenzhijiannao Capsules (60, 180, 540 mg/kg), negative control group (sterile water for injection) and positive control group (di- azepam injection by intraperitoneal injection), with 10animals in each group, all half male and female. Pentobarbital sodium were intraperitoneal injected (30±5) min after di- azepam injection by intraperitoneal injection for positive control group, and other groups were injected (90±5)min after intragastric administration. Then the areflexia number of righting reflex were recorded 30 min after administration. Re- suits There were nothing abnormal detected including general behavior, posture, gait, salivation, muscle tremors, pupils, at the time of 90 min and 24 h after administration. There were no significant difference found among each test article group and negative control group in the aspects of the number of autonomic activities, holding power and the tempera- ture (P 〉 0.05). In the nembutal subthreshold hypnotic test, the righting reflex of the positive control group was disap- peared, while the righting reflex of other groups was existed. Conclusion Under this experimental condition, there are no significant effects on the central nervous system in mice found after Shenzhijiannao Capsules single-pass intragastric administration at the doses of 60, 180, 540 mg/kg, and there is no synergy with sodium pentobarbital.
