摘要 目的研究γ干扰素(IFN-γ)对神经干细胞的增殖和自我更新能力的影响;Wnt7a蛋白在IFN-γ促神经干细胞增殖和增强自我更新能力的机制。方法从胎鼠端脑获取原代神经干细胞进行原代培养,分别添加一定浓度IFN-γ溶液和PBS溶液,分为IFN-γ处理组和PBS对照组,计算神经球体面积,细胞免疫荧光检测神经干细胞Brd U增值能力,蛋白免疫印迹检测神经干细胞IFN-γ信号通路蛋白p STAT1和Wnt7a信号通路蛋白。结果形态学显示IFN-γ增加神经球面积和Brd U+细胞数目,与对照组相比差异有统计学意义(P<0.01);蛋白免疫印迹结果显示IFN-γ促进了p STAT1和Wnt7a蛋白增加,与对照组相比差异有统计学意义(P<0.01)。结论 IFN-γ增强了神经干细胞的自我更新能力和增殖能力,并且激活了Wnt7a信号通路。 Objective To study the effect of IFN-γ on the proliferation and self-renewal of neural stem cells and the mechanism of WntTa protein in promoting the proliferation of neural stem cells. Methods Primary cultured neural stem cells were harvested from the fetal brain. The IFN-γ, and PBS solutions were added to each group. The IFN-γ group and the PBS group were used to calculate the area of the neurospheres. Detection of BrdU proliferation of neural stem cells by Cellular Immunofluorescence. Western blot was used to detect the expression of pSTAT1 and WntTa signal transduction pathway proteins in neural stem cells. Results The morphology revealed that the area of neurospheres and number of BrdU^+ cells were significantly increased by IFN-γ treatment (P〈 0.01 ) ; Western blot revealed the protein expression of STAT1 (P〈0.01) and WntTa (P〈0.01) in IFN-3, group were higher than those in PBS group, the difference was statistically significant. Conclusion IFN-γ enhanced the ability of self-renewal and proliferation of neural stem cells, and activated the WntTa signal pathway.
