斑马鱼听觉诱发电位检测系统的建立和应用 被引量:2

摘要 目的建立斑马鱼听觉诱发电位(auditory evoked potential,AEP)检测系统。方法设计并制作斑马鱼听觉诱发电位检测水箱;应用该系统检测和记录35条体长为10~46mm斑马鱼的AEP,将斑马鱼分为5组:体长12~15mm,6条;体长17~20mm,4条;体长22~26mm,4条;体长32~37mm,9条;体长42~46mm,12条。对照组为金鱼,体长38~54mm,8条。通过麻醉实验、鱼鳔实验、噪声暴露实验并以金鱼为对照验证斑马鱼AEP检测系统的可靠性。结果实验证实斑马鱼AEP检测系统获得的响应为听觉电生理反应;所有体长斑马鱼的听力频率范围为100Hz^12kHz,在发育过程中无听觉频率扩展的现象;成鱼最佳听力范围为600Hz^1kHz,5组斑马鱼AEP阈值分别为141.7±1.32、124.8±1.31、121.8±1.49、117.8±1.09和124.4±1.87dB w;斑马鱼AEP阈值随体长增加而变化,具有随着发育成熟AEP阈值降低,随着衰老阈值升高的特征。结论本研究建立了稳定的斑马鱼听觉诱发电位检测系统,该系统可用于斑马鱼AEP的检测。 Objective To establish an auditory evoked potential(AEP)detection system in zerbafish.Methods The AEP detection tank was designed and made,and then verified for its quality and reliability via four experiments:anesthesia experiment,swim bladder deflation,noise exposure and goldfish AEP test.Finally,zebrafish(total length form 10 mm to 46mm)were determined using this system for AEP.Zebrafish randomly were divided into five groups according to total length(TL=12~15mm,n=6;TL=17~20mm,n=4;TL=22~26mm,n=4;TL=32~37mm,n=9;and TL=42~46mm,n=12).Goldfish,as control group,were purchased for local petshop(TL=38~54mm,n=8).Results The results of these four verifying experiments confirmed the biological,rather than artefactual,nature of the responses represented by the recorded waveforms.The AEPs were detected up to a much higher frequency limit(12kHz)than previously reported.In this study,all fish demonstrated a range of hearing frequency from 100 Hz to 12 kHz without frequency expansion during development.The best hearing was observed at 600 Hz~1kHz.The mean values of the frequency-averaged thresholds(mean SEM)were141.7±1.32,124.8±1.31,121.8±1.49,117.8 ±1.09 and 124.4±1.87 dB w,respectively,for the 5TL groups.The AEP thresholds demonstrated both developmental improvement and age-related loss of hearing sensitivity.Conclusion An auditory evoked potential detection system of zerbafish has been established with stable performance and can be used for AEP detection of zebrafish.
