摘要
目的观察姜黄素对高糖诱导的INS-1胰岛β细胞凋亡的作用,探讨其抗糖尿病作用的可能机制。方法流式细胞术检测细胞凋亡情况;透射电子显微镜观察INS-1细胞的形态学变化;采用激光共聚焦和流式细胞术检测INS-1细胞线粒体膜电位的变化;用Western blotting法检测Bcl-2、Bax蛋白的表达量。结果姜黄素浓度为10μmol/L抗凋亡作用效果最为明显(P<0.05)。与正常组相比,模型组INS-1细胞凋亡率明显升高(P<0.05),形态发生改变,线粒体膜电位降低,Bcl-2蛋白表达明显降低(P<0.05),Bax蛋白表达明显升高(P<0.05),Bcl-2/Bax比值降低。与模型组比较,姜黄素改善凋亡的INS-1细胞的形态,提高线粒体膜电位,降低Bax蛋白表达(P<0.05),增加Bcl-2蛋白表达(P<0.05),Bcl-2/Bax比值升高。结论姜黄素改善高糖诱导的INS-1细胞凋亡状态,其抗凋亡机制与线粒体途径有关。
Objective To observe the effect of curcumin on INS-1 cell apoptosis which was induced by high glucose, and examine its mechanism. Methods Cell apoptosis was detected by flow cytometry. The morphological changes of INS-1 cells were observed by transmission electron microscope. The changes of mitochondrial membrane potential in INS-lcells were measured by confocal laser scanning microscope and flow cytometry. Western blot was used to examine the expression of Bax and Bcl-2 proteins. Results The curcumin (10ktmol/L) was effective against apoptosis (P〈0.05). Compared to the normal group, apoptosis rate of INS-1 cells was significantly increased in the model group (P〈0.05). The morphology was changed, while mitochondrial membrane potential and expression of Bcl-2 protein were decreased (P〈0.05), and the expression of Bax protein was significantly increased (P〈0.05). There were some effects of curcumin on INS-1 cells, such as ameliorating cell apoptotic morphology, significantly increasing the mitochondrial membrane potential, reducing the protein expression of Bax and increasing the protein expression of Bcl-2. Conclusion Curcumin may positively affect the apoptosis of INS-I cells exposed to high glucose. The mechanism is associated with the mitochondrial pathway.
