养正消积胶囊对荷MFC胃癌小鼠调节性T细胞的影响 被引量:4

摘要 目的探讨养正消积胶囊对荷MFC胃癌小鼠的疗效及免疫调节机制。方法雄性615小鼠50只,皮下接种法建立小鼠胃癌肿瘤模型,随机分为模型组、养正消积胶囊组、5-氟脲密啶(5FU)组、联合组,每组各10只,另设正常组10只。观察荷MFC胃癌小鼠移植瘤生长情况以及体质量等的变化。于末次给药后完整剥取移植瘤、胸腺、脾脏并称重,计算各组荷MFC胃癌小鼠移植瘤体积和抑瘤率。流式细胞仪测定外周血CD4^+CD25^+Foxp3^+Treg细胞比例。结果 (1)模型组、联合组、养正消积胶囊组、5FU组小鼠瘤重分别为(4.46±0.89)g、(2.21±0.27)g、(3.50±0.42)g、(3.30±0.41)g,与模型组瘤重比较,联合组、养正消积胶囊组、5FU组抑瘤率分别为50.92%、22.29%、25.89%,差异均有统计学意义(P<0.05)。(2)联合组、养正消积胶囊组小鼠外周血CD4^+CD25^+Foxp3^+Treg细胞比例分别为(4.73±0.35)%、(5.43±0.47)%,均低于模型组的(7.26±1.12)%,差异有统计学意义(P<0.05)。结论养正消积胶囊具有抗MFC胃癌移植小鼠肿瘤生长的作用,该作用可能与其下调CD4^+CD25^+Foxp3^+Treg细胞表达相关。 Objeetive To investigate the anti-tumor effects ofYangzheng Xiaoji Capsule and theunderlying im- mune regulation mechanism. Methods Forty of MFC tumor bearing 615 mice models were established by inoculated subcutaneously way, then were randomly divided into 4 groups (n=10 in each group): model group, Yangzheng Xiaoji Capsule group, 5FU group, and comprehensive treatment group. Another 10 mice served as control group. The growth of tumor in mice and the change of mouse weight were observed. After last dosing, the mice were sacrificed to obtain and weigh the tumor, thymus and spleen, then the tumor volume and the inhibitory rate were calculated. CD4+CD25+ Foxp3+Treg cells proportion in mouse peripheral blood was detected with Flow cytometry. Results The tumor weight in model group, comprehensive group, Yangzheng Xiaoji Capsule group, and 5FU group were 4.46±0.89g, 2,21±0.27g, 3,50±0.42g, and 3.30±0.41g, respectively. Compared to model group, the inhibitory rateof comprehensive treatment group, Yangzheng Xiaoji Capsule group, and 5FU group were 50.92%, 22.29%, and 25.89%, respectively, with significant difference(P〈0.05). The expression of CD4+CD25+Foxp3+Tregcellsin comprehensive treatment group and Yangzheng Xiaoji Capsule group waslower than that of model group (4.73%±0.35%, 5.43%±0.47% vs 7.26%±1.12%, P〈0.05). Conclusion Yangzheng Xiaoji Capsule can suppress the growth of MFC gastric carcinoma, which may be involved in decreasing the level of CD4+CD25+Foxp3+Treg.
