补肾健脾中药复方对骨质疏松骨折骨痂中CTGF、Osterix因子表达的影响

Effect of Chinese Herbal Compound of Tonifying Kidney and Strengthening Spleen on CTGF and Osterix factor expression in callus of osteoporotic fracture

目的分析补肾健脾中药复方对骨质疏松骨折骨痂中CTGF、Osterix因子表达的影响。方法选择6月龄清洁级健康雌性SD大鼠60只,随机分为3组:模型组、西药组和中药组,每组20只。骨质疏松骨折模型成功后,中药组和西药组分别给予补肾健脾中药复方和阿伦磷酸钠混悬液灌服,模型组进行常规培养,1个月后取各组骨折的骨痂,进行体外培养,各组骨痂给予对应含药血清进行培养。采用荧光定量PCR法检测各组CTGF、Osterix因子的表达。结果在CTGF表达方面,中药组和西药组中的CTGF m RNA含量明显高于模型组,差异有统计学意义(P<0.05);在Osterix表达方面,中药组和西药组中的Osterix m RNA含量明显高于模型组,差异有统计学意义(P<0.05),且中药组的Osterix m RNA含量高于西药组(P<0.05)。结论补肾健脾中药复方能在一定程度上促进骨痂的生长,从而促进骨折愈合,为骨质疏松性骨折愈合的机制和防治方法提供了一种新的途径。

Objective To analyze the influence of Chinese Herbal Compound of Tonifying Kidney and Strengthening Spleen on CTGF and Osterix factor expression in callus of osteoporotic fracture.Methods 60 female SD rats of cleanclass aged six-month were selected and they were randomly divided into 3 groups：model group,western medicine group and traditional Chinese medicine group （TCM group） （n=20）.After the success of the osteoporosis＇ fracture model,TCM group was given Chinese Herbal Compound of Tonifying Kidney and Strengthening Spleen and the western medicine group was given sodium alendate suspension.The model group was given routine culture.After 1 month,the callus of each group was taken and cultured in vitro.The callus of each group was cultured with the corresponding serum.CTGF and Osterix expression in each group were detected by fluorescence quantitative PCR.Results The expression of CTGF mRNA in TCM group and western medicine group was significantly higher than that in model group （P〈0.O5）.Osterix mRNA level in TCM group and western medicine group were significantly higher than those in model group （P〈0.05）,and the content of Osterix mRNA in TCM group was higher than that in western medicine group （P〈0.05）.Conclusion Chinese herbal Compound of Tonifying Kidney and Strengthening Spleen can promote the growth of callus to a certain extent,thus pro- mote fracture healing,and it provides the new way for the mechanism and prevention and treatment methods osteoporosis＇fracture healing.
