摘要
目的探讨各剂量黄连解毒汤不同给药时长对营养性肥胖大鼠的毒性。方法雄性SD大鼠100只,按体质量随机分为正常对照组、模型对照组、黄连解毒汤高、中、低剂量组,每组20只,每3日测量一次体质量及摄食量。除正常组外,每组均采用三重喂养法复制大鼠模型。给药22天与42天后各处理10只动物,计算脂肪指数,检测血清Alb、TP、TBIL、GGT、ALT、ALP、Cre和CYP450含量。结果黄连解毒汤能有效降低营养性肥胖大鼠的体质量及脂肪指数,升高血清ALT、GGT、TBIL,降低血清Alb、TP及CYP450,以上结果具有统计学意义(P<0.05或P<0.01)并与摄食量减少无明显相关性。结论黄连解毒汤可能对营养性肥胖大鼠产生肝、肾毒性并抑制代谢活性,临床应用时建议综合考虑其对机体的影响。
Objective To investigate the toxicity of Huanglian Jiedu decoction with different dosage and administration time on rats with nutritional obesity. Methods 100 male SD rats were randomly divided into normal group, model group, low dose group, medium dose group and high dose group, with 20 in each group. In addition to the normal group, the rest of the groups were to be made a model of rats with nutritional obesity by using the "high fat emulsion, sodas and ordinary diet" way for 3 weeks. The administration time after model establishment was 22 days and 42 days. 10 rats were to be sacrificed for each time. After the last administration, rats were executed under anesthesia to separate serum and strip fat tissue around genitals and kidneys. Finally, we inspected the content of Alb, TP, TBIL, GGT, ALT, ALP, Cre and CYP450, and calculated the fat index. Results Huanglian Jiedu decoction can effectively reduce the weight, fat index, Alb, TP and CYP450 level of obese rats, and increase the serum level of ALT, GGT and TBIL. The above results were statistically significant (P 〈 0.05 or P 〈 0.01 ) , and no significant correlation was found with the decrease of food intake. Conclusions It is possible for Huanglian Jiedu decoction to produce liver and kidney toxicity and inhibit metabolic activity in nutritional obese rats. Comprehensive reference of HLJDT on various indexes is recommended in clinical applications.
