摘要
PTEN基因是一个具有双重特异磷酸酶活性的抑癌基因,可影响细胞周期调控和细胞增殖.异常PTEN可导致PI3K通路失调,细胞异常增殖.多种肿瘤中可检测到异常的PTEN突变.最近的研究显示,除了在PI3K通路中的作用外,PTEN还具有DNA损伤修复等其它生物学功能,PTEN缺陷肿瘤细胞对DNA损伤药物敏感.因此,针对PTEN基因异常肿瘤可采取PI3K抑制剂、DNA损伤化疗药物等治疗方法联合使用的个体化治疗策略,本文对人PTEN基因异常肿瘤的个体化治疗进行初步分析.
PTEN is a tumor suppressor gene characterizing with dual phosphatase activities, which can affect cell cycle regulation and cell proliferation. Aberrant PTEN results in dis-regulation of PDK cascade pathway and abnormal cell proliferation. Coincidently, PTEN mutants were detectable in various types of cancers. Besides of its effect in PI3K pathway, recent reports demonstrate PTEN also functions in DNA repair and other biological activities ; while PTEN deficient tumor cells are sensitive to DNA damage drugs. Thus, PI3K inhibitors and DNA damage drugs can be combined to target tumor cells with different PTEN mutants, which would be a promising personalized therapy. This paper primarily analyzes the individual therapeutic strategies to tumors with PTEN mutants.
