摘要
目的研究精子DNA完整性不同检测方法的区别,探讨使用精子染色质结构分析法(SCSA)和精子染色质扩散实验法(SCD)分析精子DNA碎片化率(DFI)结果的一致性。方法分别用SCSA法和SCD法检测119例不育症患者精液的DFI值,采用SPSS 22.0软件进行相关性分析和配对t检验。结果 SCD法检测119例精液标本DFI均值为(15.4±8.9)%,SCSA法DFI均值为(14.2±7.3)%SCD法DFI值高于SCSA法,差异有统计学意义(P<0.05)。SCSA法和SCD法相关性分析呈明显的线性正相关(r=0.816,P<0.01)。结论 SCSA法与SCD法比较,其检测精子DFI结果具有一致性;但SCD法检测值普遍比SCSA法高。
Objective To assess the difference of different methods in the detection of sperm DNA integrity, and to discuss the consistence of using sperm ehromatin structure assay ( SCSA ) and sperm chromatin dispersion test ( SCD ) in the detection of DNA fragmentation evaluation. Methods We used SCSA and SCD assays to detect DNA fragmentation in sperm from 119 infertile men. The correlation analysis and t - test were performed with SPSS 22.0 software. Results The mean DFI value was ( 15.4 ± 8.9) % by SCD and ( 14.2 ± 7.3) % by SCSA in 119 sperm samples. The SCSA assay was strongly correlated with the SCD assay for sperm DNA fragmentation( r = 0. 816, P 〈 0.01 ). However, the mean DIF values with SCD was higher than that with SCSA, with the difference statistically significant(P 〈 0.05 ). Conclusion There is a strong positive correlation between SCSA and SCD assays in tile detection of DNA fragmentation. SCD assay showed higher levels of DNA fragmentation than that measured by SCSA.
