摘要 为优化黄花蒿(Artemisia annua Linn)离体再生体系,并建立黄花蒿遗传转化途径,选用黄花蒿常规品种428-A为材料,以子叶、幼嫩叶片、顶芽作为外植体,设计不同植物生长调节剂组合的培养基配方。结果表明,黄花蒿子叶适宜作为组织培养的外植体,MS+1.5 mg/L 6-BA+0.05 mg/L NAA培养基有利于诱导愈伤组织形成和分化,其出愈率及出芽率分别高达89.0%、85.7%,1/2MS+0.05 mg/L NAA+0.05mg/L IAA培养基有利于再生植株不定根的诱导,生根诱导率达94.0%。 In order to optimize Artemisia annua Linn in vitro regeneration system and establish transformation way,the con-ventional varieties 428-A of Artemisia annua Linn as material was chose. Cotyledon,young leaves, crown were used as ex-plant,the simple and effective regeneration system of Artemisia annua Linn was set up,by using different types and different concentrations of plant growth regulator of the formula. The results indicated that the young leaves is the best material for the tissue culture of explant. The best solid medium of callus formation and differentiation is MS+1.5 mg/L 6-BA+0.05 mg∕L NAA,the rate of callus formation is 89.0% and the rate of budding is 85.7%. The most useful solid medium of inducing of root is 1∕2MS+0.05 mg∕L NAA+0.05 mg∕L IAA, in which the rooting rate reached 94.0%.
