摘要
目的研究miRNA-21在膀胱癌中对PTEN的调控作用。方法采用双荧光素酶报告基因实验与Western Blot实验检测miRNA-21对膀胱癌HTB-9细胞中PTEN表达的影响。结果 miRNA-21可负向调控PTEN的表达;转染miRNA-21反义核苷酸、沉默miRNA-21后,膀胱癌HTB-9细胞中PTEN的表达增加。结论 miRNA-21可通过调控膀胱癌中PTEN的表达影响膀胱癌的发生与进展。
Objective To study the regulatory effect of miRNA-21 on PTEN in bladder cancer. Methods The miRNA-21 expression was directly regulated by PTEN through fluorescence report experiments. The expression of PTEN in HTB-9 was detected by Western blot after transfection with miRNA-21. Results The miRNA-21 negatively regulated the expression of PTEN and increased the expression of PTEN in bladder cancer cell line HTB-9 after miRNA-21 silencing. Conclusion The miRNA-21 can regulate the expression of PTEN in bladder cancer,which affects the occurrence and development of bladder cancer.
