左乙拉西坦对难治性癫痫大鼠海马组织中Slit2表达的影响 被引量:1

摘要 目的:观察左乙拉西坦对难治性癫痫大鼠海马组织中Slit2表达的影响。方法:Wistar大鼠随机分为对照组,模型组(IE组),治疗组(LEV治疗组),LEV对照组(LEV组)。腹腔注射氯化锂-匹罗卡品建立难治性癫痫大鼠模型,造模成功后LEV组和LEV治疗组给予LEV 200mg/(kg·d)灌胃,对照组和IE组给予等量生理盐水,应用免疫组化法在第1周、2周和4周检测各组海马组织中Slit2的表达量。结果:LEV组和对照组比较,Slit2的表达量差异无统计学意义(P>0.05);IE组Slit2表达量明显降低,与对照组比较差异有统计学意义(P<0.05);LEV治疗组与IE组比较,Slit2表达量明显升高,差异有统计学意义(P<0.05)。结论:Slit2的表达变化促进海马发生苔藓纤维化(MFS)参与难治性癫痫形成,LEV控制癫痫发生,可能与上调Slit2的表达有关。 Objective: To observe the effect of levetiracetam (LEV) on Slit2 in hippocampus of rats with frac- tory epilepsy. Methods: Wistar rats were randomly divided into control group, model group (IE group) , treatment group (LEV treatment group), and LEV control group (LEV group). Refractory epilepsy rat modes were induced by intraperitoneal injection of lithium - pilocarpine ; LEV group and LEV treatment group were given levetiracetam (200mg/kg per a day) by filling the stomach after the mode was established successfully; the control group and IE group were given the same dose of saline solution. The expression of Slit2 in rat hippoeampal's tissues were detected respectively at the 1st week, 2nd week and 4th week by immunohistochemical method. Results: There was no sig- nificant difference between LEV group and control group (P 〉 0.05). Compared with the control group, the expression of Slit2 of IE group decreased significantly (P 〈 0.05 ). Compared with IE group, the expression of Slit2 of LEV treatment group increased significantly (P 〈 0.05 ). Conclusion: The expression of Slit2 promotes rat hippocampal mossy fiber sprouting(MFS) involving in the formation of refractory epilepsy. Levetiracetam is correlated with raising expression Of Slit2 and redueing MFS when it controls the incidence of seizures during curing the refractory epilepsy.
