慢性粒细胞白血病相关的癌基因、抑癌基因表达谱的研究

摘要 运用表达谱芯片技术对慢性粒细胞白血病相关的癌基因、抑癌基因表达谱进行研究 对慢性粒细胞白血病及正常对照细胞进行总RNA抽提 ,纯化后的mRNA进行逆转录制备杂交探针 ,应用Biostar 10 2 4条人类全长基因cDNA芯片筛选与CML相关的基因 杂交信号用ScanArray 5 0 0 0扫描仪扫描 ,用ImaGene 3.0软件分析 ,发现有9个癌基因、4个抑癌基因同时在慢性粒细胞白血病异常表达 分析这些异常基因表达谱 。 In order to understand the molecular pathophysiology of chronic myeloid leukemia, differently expressed genes in chronic myeloid leukemia (CML) were investigated and some CML associated oncogenes,tumor suppressor genes were determined by cDNA microarray.The mRNA from 3 cases of CML and normal cells were reversely transcribed to cDNA with the incorporation of fluorescent labeled dUTP to prepare the hybridization probes,respectively.After hybridization biostar 1024 cDNA microarray were scanned for the fluorescent intencity.CML associated oncogenes,tumor suppress genes were screened through the analysis of difference in gene expression profile.Among 1024 target genes,there were 9 oncogenes,4 tomor suppressor genes whose expression level differed from normal and CML cells.Analysis of there differentially expressed oncogenes,tumor suppressor genes will be helpful for further undersdanding the molecular mechanism of CML.
