一种烟草蛋白激酶基因表达的原位定位分析

摘要 将玉米MCK1同源的烟草NtCaMK3cDNA的 3′非翻译区 (3′UTR)构建到pBluescriptⅡSK(- )载体中 ,并以此 3′非翻译区为模板经体外转录得到了地高辛标记的反义探针用于原位杂交 ,研究结果表明该基因在烟草的雄性和雌性生殖器官及胚中均有表达 ,且在不同生长发育阶段其表达部位有较大差异 ,证明该基因表达受发育的调控 ,并具有一定的组织细胞分布特性 ,显示该基因可能参与了Ca2 + The 3′untranslated region (3′UTR) of NtCaMK3,a homolog of maize MCK1, was subcloned into the Xho Ⅰ site of pBluescript SK(-). An antisense probe transcripted from the 3′UTR in vitro was used for the location of NtCaMK3′s expression. Our results indicated that the expression of NtCaMK3 is developmental\| and tissue\|specific both in tobacco male and female reproductive organ, suggesting that Ca 2+ /CaM signal transduction system mediated by NtCaMK3 play a role in plant growth and development.
