摘要 目的研究Siglec—E在脓毒症小鼠RAW264.7单核巨噬细胞的表达。方法培养小鼠单核巨噬细胞株RAW264.7,取对数生长期细胞,分别为对照组(Con组)、实验组(LPS组)在不同的时间点提取总的RNA和总蛋白,用RT—PCR法测细胞裂解液中Siglec—EmRNA表达;用Western blot测胞质Siglec—E蛋白含量。结果不同浓度的脂多糖(LPS)刺激后6~12h培养细胞Siglec—EmRNA和蛋白表达相对于Con组增高,24h表达量显著增高,而24~48h含量逐渐减少,各不同时间点的差异有统计学意义(P〈0.01)。结论Siglec—E在脓毒症炎症时表达量增加且呈现一定的时间和剂量依赖。 Objective To investigate the expression of siglec -E in monocytes and macrophages of RAW 264.7 in septic mice. Methods RAW 264.7 cells in logarithmic growth phase were separated into blank control group and lipopolysaccharide (LPS) group. After LPS intervention, total RNA and protein were extracted to determine the mRNA and protein expression of siglec - E in monocytes and macrophages of RAW 264.7 in septic mice by the method of RT - QPCR and Western blot, respectively. Results Stimulation of LPS up - regulated the mRNA and protein expression of siglec - E both time - dependently and dose - dependently. Conclusion Expression of siglec - E in monocytes and macrophages in septic mice is high, and in a time - dependent and dose - dependent manner to some extent,
