摘要
根据槲皮素能与铝离子形成稳定的荧光络合物,建立同步荧光法测定荷叶中总黄酮含量的方法。以槲皮素为标样,选择发射波长与激发波长的波长差△λ为40 nm扫描同步荧光光谱,测定荷叶中总黄酮含量,并进行加标回收试验验证方法的可靠性。试验结果表明,槲皮素浓度在0.008μg/mL^0.40μg/mL范围内与荧光强度具有良好的线性关系,线性回归方程:y=5 780x+3.27,相关系数R为0.999 0。
On the basis of reaction of quercetin with Alumimum ion to form fluorescent material, a new method of determining total flavonoids in lotus leaves was established. The content of total flavonoids in lotus leaves was determined, taking quercetin as standard sample, selecting△λ=40 nm and scanning by synchronous fluorom-etry. Then the accuracy was inspected by recovery test. The experimental results showed that when the quercetin concentration ranged from 0.008 μg/mL to 0.40 μg/mL, fluorescence intensity and quercetin concentration showed the good linear relationship,the linearity relationship was y=5780x+3.27, the correlation coefficient R was 0.9990.
