莲子红衣多糖对胰脂肪酶活性抑制作用的研究 被引量:7

摘要 为考察莲子红衣多糖对体外胰脂肪酶活性的抑制效果,采用分光光度法测定了不同条件下对胰脂肪酶活性抑制作用的影响,并在单因素试验基础上运用L_9(3~4)正交法优化得到莲子红衣多糖对胰脂肪酶活性抑制作用的最佳条件。结果显示,在pH 7.2、莲子红衣多糖提取物浓度0.15 g/mL、反应时间10 min条件下,可达到最高抑制率94.61%。此外,Dixon和Lineweaver-Burk分析表明,莲子红衣多糖对胰脂肪酶的抑制作用类型为非竞争性抑制,抑制常数为0.073 6 g/mL。 The inhibitory effect of polysaccharide from lotus seed skin on pancreatic lipase was discussed. The pancreatic lipase activity in different conditions was measured by using spectrophotometry. On the basis of single factor experiment, the inhibitory conditions were optimized by L9 (3^4) orthogonal method. The results revealed that the optimal inhibition rate was 94.61% under the conditions of pH 7.2, polysaccharide concentration 0.15 g/mL and reaction time 5 min. Dixon and Lineweaver-Burk proved that polysaccharide from lotus seed skin was a non-competitive type of inhibitor on pancreatic lipase, and its inhibition constant was 0.073 6 g/mL.
