穿心莲种子无菌萌发条件优化研究 被引量:1

摘要 目的优化穿心莲种子无菌萌发的实验条件。方法以植物组织培养技术,采用单因素法考察采收时间、种子的成熟状态、光照条件、预处理、消毒时间、培养基对穿心莲种子萌发的影响。结果穿心莲种子萌发的优化条件为:贮藏时间半年、完全成熟的种子、有光照条件、40%硫酸浸泡2 h预处理、消毒5 min,培养基为MS或1/2 MS。结论采用该优化条件可以在短期内培育大量整齐一致的穿心莲试管苗,且不受季节和环境条件的限制,具有较强的实用性和推广性。 Objective To optimize the experimental conditions for aseptic germination of Andrographis paniculat, (Burm. f.) Nees. seeds. Methods Plant tissue-culture technique was applied for the investigation of time after seed harvesting, seed maturity, illumination intensity, pretreatment, disinfection time and culture medium on the germination of Andrographis paniculata (Burro. f.) Nees. seeds by single factor way. Results The optimal conditions for the germination of Andrographis paniculata (Burro. f.) Nees. seeds were as follows: half a year after scrod harvesting, mature seeds, light condition, soaking the seeds in 40 % H2SO4 for 2 h, disinfection for 5 min, cuhuring in MS or 1/2 MS medium. Conclusion Under the optimized conditions, a large number of neat test-tube seedlings were obtained after cultivating for short time without seasonal and environmental restrictions, indicating that the established method is practical and worthy of the promotion.
