摘要
目的研究毛蕊异黄酮苷在大鼠血清中的相关代谢产物。方法甲醇沉蛋白法进行样品前处理后,采用超高效液相-电喷雾-四级杆串联飞行时间质谱(UPLC/Q-TOF-MS)在正离子模式下检测大鼠口服毛蕊异黄酮苷1h后血清中的相关代谢产物。结果共鉴定出了5个毛蕊异黄酮苷在大鼠体内相关代谢物,分别为葡萄糖醛酸化的毛蕊异黄酮苷(M2)、葡萄糖醛酸化的毛蕊异黄酮(M3)、毛蕊异黄酮(M4)和硫酸化的毛蕊异黄酮(M5和M6)。结论该方法对毛蕊异黄酮苷吸收入血的相关代谢成分进行了初步研究,为后续研究其在体内发挥药效的活性成分形式提供了实验基础。
Objective To study the related metabolites of calycosin-7-O-β-D-glucoside in rat serum. Method The samples obtained in rats after lh oral administration calycosin-7-O-β-D-glucoside were pretreated with methanol and analyzed by ultra-high performance liquid-electrospray-quadrupole combined time-of-flightmass spectrometry (UPLC/Q-TOF-MS) in positive ion mode. Results Five metabolites of calycosin-7-O-β-D-glucoside in the rat serum were identified, the five metabolites are calycosin-7-O-β-D-glucoside-3'-O-glucuronide(M2), calycosin-7-O-β-D-glucuronide(M3 ), calycosin(M4), calycosin-7-O-sulfate(M5) and calycosin-4'-O-sulfate(M6), respectively. Conclusion This method has been used to study the metabolites of calycosin-7-O-β-D-glucoside in rat serum initially, which provides an experimental basis for the subsequent study of its active constituents in vivo.
