摘要
目的揭示Ras家族小G蛋白——RASD1在胶质瘤U251细胞迁移中的作用。方法在稳定过表达RASD1的胶质瘤U251细胞系中,应用Western blot验证RASD1过表达效果;应用鬼笔环肽染色显示细胞骨架蛋白F-actin的分布;应用细胞划痕实验检测过表达RASD1对胶质瘤U251细胞迁移的影响。结果胶质瘤U251细胞系中外源性RASD1过表达成功。过表达RASD1降低了细胞骨架蛋白F-actin在细胞板状伪足、侵袭伪足上分布。稳定过表达RASD1的胶质瘤U251细胞迁移能力下降。结论 RASD1具有抑制胶质瘤U251细胞迁移能力的作用。
Objective To reveal the role of dexamethasone-induced Ras-related protein 1(RASD1),one of the Ras family small G proteins,in the migration of glioma U251 cell. Methods Western blot was performed on the U251 cell line to verify the overexpression of RASD1;PhalloidinF-actin immunofluorescence was performed to observe the distribution of cytoskeletal protein F-actin;and cell wound scratch assay was performed to evaluate the effect of RASD1-overexpression on the migration of U251 cells. Results Exogenous RASD1 was successfully overexpressed in U251 cells,which reduced the accumulation of F-actin in the lamellipodiaand invasion-filopodia,and inhibited the migratory ability of U251 cells. Conclusion RASD1 may inhibit the migration of glioma U251 cells.
