宁夏密点麻蜥对胃癌前病变模型大鼠HIF-α、VEGF的影响 被引量:3

摘要 目的:研究不同剂量宁夏密点麻蜥对胃癌前病变(PLGC)模型大鼠缺氧诱导因子HIF-1α和血管内皮生长因子VEGF的影响,探讨宁夏密点麻蜥对PLGC的调控机制。方法:60只SPF级SD雄性大鼠,随机分为空白对照组、PLGC模型组、宁夏密点麻蜥高、中、低剂量组、维酶素对照组。除空白对照组外,其余各组均以0.02 mmol/L的MNNG,配合40%乙醇、饥饱失常法等饲养24周,制备PLGC大鼠模型。分别以宁夏密点麻蜥高、中、低剂量,维酶素连续治疗12周,观察PLGC模型大鼠胃黏膜病理变化;应用免疫组化法检测胃体组织HIF-1α和VEGF表达水平。结果:通过Image-Pro Plus6.0图像分析系统进行图像光密度测算,各模型组与空白对照组HIF-1α和VEGF水平比较,AOD值明显降低(P<0.05);各治疗组与模型组HIF-1α和VEGF水平比较,AOD值明显降低(P<0.05);宁夏密点麻蜥高、中、低剂量治疗组HIF-1α和VEGF均低于维酶素组(P<0.05),其中以高剂量组AOD值最小。结论:宁夏密点麻蜥能抑制PLGC模型大鼠HIF-1α和VEGF表达,逆转胃黏膜病理改变,抑制癌变趋势。 Objective : To research the influence of different doses of Eremias multiocellata from Ningxia on Hy- poxia inducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF) in model rat with gastric precancerous lesions (PLGC) , and discuss the regulation mechanism of Eremias multiocellata from Ningxia on PLGC. Methods: 60 SD male rats of SPF grade were randomly divided into blank control group, PLGC model group, high, medium and low dose groups of Eremias multiocellata from Ningxia, vitacoenzyme control group. Except blank control group, all the rest groups were prepared into PLGC rat models in 12 weeks with O. 02 mmol/L MNNG,combined with 40% ethanol and abnormal starvation and satiety etc. After the continuous treatment for 12 weeks respectively with high, medium and low dose of Eremias multiocellata from Ningxia and vitacoenzyme, the pathological change of gastric mucosa in PLGC rat models was observed; HIF-1α and VEGF expression of gastric tissue was detected by immunohistochemistry. Results: The image optical density was measured and calculated by Image-Pro Plus6.0 image analysis system. HIF-1α and VEGF levels of each model group and blank control group were compared,AOD value was significantly lowered( P 〈0.05 ). HiF-1α and VEGF levels of each treatment group and model group were compared, AOD value was significantly lowered(P 〈 0.05 ). HIF-1α and VEGF lev- els of high, medium and low dose treatment groups of Eremias multiocellata from Ningxia were lower than those of vitacoenzyme group ( P 〈 0.05 ) ; among them, the AOD value of high dose group was the smallest. Conclusion : Eremias multiocellata from Ningxia can suppress HIF- 1αand VEGF expression in PLGC rat model, reverse the pathological change of gastric mucosa and suppress the trend of canceration.
