摘要
目的探究硫化氢(H2S)对糖尿病小鼠胰岛素抵抗的作用和可能的分子机制。方法 C57Bl/6J小鼠随机分为正常对照组、糖尿病组、低剂量治疗组、高剂量治疗组及阳性对照组,四氧嘧啶联合高脂饮食复制糖尿病小鼠模型,检测各组小鼠脂肪组织中Caspase-3的活性、B淋巴细胞瘤-2基因(Bcl2)、Bcl-2相关X(Bax)蛋白的表达水平,以及空腹血糖、空腹血清胰岛素和血浆H2S浓度,并计算胰岛素敏感指数。结果糖尿病小鼠Caspase-3活性和Bax表达水平下调,而Bcl2蛋白表达水平上调,治疗组各指标得到缓解。结论 H2S可以减轻胰岛素抵抗,其机制可能为减轻脂肪组织的细胞凋亡。
Objective To investigate the effect of hydrogen sulfide on insulin resistance and its mechanism in mice with diabetes mellitus. Methods C57B16/J mice were randomly divided into normal control group, dia- betes mellitus group, low-dose therapy group, high-dose therapy group and positive control group. Diabetes mellitus models were established by high-fat diet and tetraoxypyrimidine. Caspase-3 activity, Bcl-2 and Bax expressions in adipose tissue were measured, as well as fasting plasma glucose, fasting serum insulin and hy- drogen sulfide in plasma. Then, insulin sensitivity index was calculated. Results Caspase-3 activity and Bax level were clown-regulated, while Bcl2 was up-regulated in mice with diabetes mellitus, and all the indexes recovered after therapy with sodium hydrosulfide. Conclusions Hydrogen sulfide could alleviate insulin resis- tance by reducing the apoptosis of adipose cells.
