微小RNA-100与微小RNA-143在膀胱癌患者血清的表达及临床意义 被引量:1

摘要 目的探讨联合检测血清微小RNA(miRNA,miR)-100与miR-143水平对膀胱癌的诊断价值。方法应用实时定量反转录聚合酶链反应(RT-qPCR)检测112例膀胱癌患者和100例健康受试者血清中miR-100与miR-143的水平,并应用受试者工作特征(ROC)曲线计算miR-100与miR-143的临界值,并根据此临界值单独或联合检测血清中miR-100与miR-143水平,观察其诊断膀胱癌的灵敏度与特异度。结果血清miR-100相对表达水平在NMIBC组(0.213±0.087)和MIBC组(0.175±0.101)分别显著低于对照组(1.086±0.493;χ2=7.381,P=0.026;χ2=12.322,P=0.011);血清miR-143相对表达水平在NMIBC组(0.193±0.075)和MIBC组(0.011±0.003)分别显著低于对照组(0.992±0.334;χ2=9.557,P= 0.019;χ2=16.926,P=0.008);MIBC组血清miR-143相对表达水平(0.011±0.003)显著低于NMIBC组(0.193±0.075;χ2=4.346,P=0.033)。存在淋巴结转移的膀胱癌患者血清中miR-100与miR-143的相对表达水平(0.231±0.027、0.224±0.117)分别显著高于无淋巴结转移的膀胱癌患者(0.144±0.065、0.162±0.044;χ2=5.861,P=0.028;χ2=3.793,P=0.031);血清miR-100与miR-143联合检测对诊断膀胱癌的灵敏度与特异度分别为89.73%与88.91%。结论膀胱癌患者血清中miR-100与miR-143水平显著降低;血清miR-100与miR-143联合检测有助于诊断膀胱癌。 ObjectiveTo observe the value of combined detection of the level of microRNA (miRNA, miR)-100 and miR-143 in serum of bladder cancer.MethodsSerum of 112 patients with bladder cancer and 100 cases of healthy people was collected, and real-time reverse transcriptase-polymerase chain reaction(RT-qPCR) was used to detect the level of miR-100 and miR-143. The receiver operating characteristic (ROC) curve was used to calculate the cut-off value of miR-100 and miR-143, and the value of combined detection of the level of miR-100 and miR-143 in serum of bladder cancer was assessed.ResultsThe relative expression levels of miR-100 in serum in NMIBC group (0.213±0.087) and muscle invasive bladder cancer (MIBC) group (0.175±0.101) were significantly lower than the control group (1.086±0.493; χ2=7.381, P=0.026; χ2=12.322, P=0.011); the relative expression levels of miR-143 in serum in NMIBC group (0.193±0.075) and MIBC group (0.011±0.003) were significantly lower than the control group (0.992±0.334; χ2=4.346, P= 0.033); the relative expression levels of miR-143 in serum in MIBC group(0.011±0.003) was significantly lower than the NMIBC group (0.193±0.075; χ2=4.346, P=0.033). The relative expression levels of miR-100 and miR-143 (0.231±0.027, 0.224±0.117) were significantly higher than those without lymph node metastasis in patients with bladder cancer (0.144±0.065, 0.162±0.044; χ2=5.861, P=0.028; χ2=3.793, P=0.031). The sensitivity and specificity of combined detection of miR-100 with miR-143 in the diagnosis of bladder cancer were 89.73% and 88.91% respectively.ConclusionLevels of miR-100 and miR-143 in serum of bladder cancer patients are reduced significantly. There is a high diagnostic value of combed detection of miR-100 with miR-143 to bladder cancer.
