非受体型蛋白酪氨酸磷酸酶在类风湿性关节炎患者外周血单个核细胞中的表达

摘要 目的探讨非受体型蛋白酪氨酸磷酸酶(PTPN)在类风湿性关节炎(RA)患者中的表达及其临床意义。方法选取本院住院RA患者32例和健康对照29例,抽取外周血,Ficoll密度梯度离心法分离单个核细胞。利用实时定量反转录聚合酶链反应(RT-qPCR)和Western blot法检测蛋白酪氨酸磷酸酶(PTPs) mRNA及蛋白的表达水平。同时检测外周血炎性因子白细胞介素(IL)-1β、IL-6、肿瘤坏死因子(TNF)-α及生化指标红细胞沉降率(ESR)、C反应蛋白(CRP)的水平,并计算病情活动性指标DAS28评分,统计学分析PTPs的表达水平与炎性因子、生化指标、DAS28评分间的关系。结果与对照组比较,RA患者PTPN1、PTPN2、PTPN3、PTPN4、PTPN7、PTPN9、PTPN13的表达量下降,PTPN11(2.530±0.743,对照组1.000±0.034)和PTPN14(2.269±0.548,对照组1.000±0.057)表达明显上升。PTPN11的表达水平与TNF-α水平[(5.95±1.57) ng/ml,r=0.669,P=0.000]、CRP含量[(28.58±6.72) mg/L,r=0.795,P=0.000]、DAS28评分[(4.20±0.73)分,r=0.703,P=0.000]间具有显著正相关。PTPN14表达水平与TNF-α水平(r=0.633,P=0.000)和CRP含量(r=0.435,P=0.013)明显相关,但与DAS28评分(r=0.033,P=0.856)之间无明显相关。结论PTPs在RA患者外周血单个核细胞中差异化表达,其中PTPN11与RA炎性因子、CRP含量和DAS28评分呈正相关。 ObjectiveTo study the expression and clinical importance of non-receptor protein tyrosine phosphatases (PTPN) in patients with rheumatoid arthritis (RA).MethodsThirty-two RA patients enrolled in our hospital from February 2015 to October 2015 and 29 healthy controls were recruited. Peripheral blood was obtained and mononuclear cells were prepared using Ficoll density gradient centrifugation. The expression of protein tyrosine phosphatases (PTPs) mRNA and protein was detected by real-time quantitative reverse transcriptase-polymerase chain reaction (RT-qPCR) and immunoblot respectively. Meanwhile, the levels of inflammatory factors like interleukin (IL)-1β, IL-6, tumor necrosis factor (TNF)-α and biochemical indexes like erythrocyte sedimentation rate (ESR) and C-reactive protein (CRP) were evaluated, with the calculation of DAS28, a measure of disease activity in RA. The correlation between PTPs and inflammatory factors, or biochemical indexes, or DAS28 was analyzed.ResultsIn comparison with the controls, the expression of PTPN1, PTPN2, PTPN3, PTPN4, PTPN7, PTPN9, PTPN13 was reduced, while that of PTPN11 (2.530±0.743 vs. 1.000±0.034) and PTPN14 (2.269±0.548 vs. 1.000±0.057) was augmented in RA patients. The expression of PTPN11 was positively correlated with TNF-α levels [(5.95±1.57) ng/ml, r=0.669, P=0.000], CRP levels [(28.58±6.72) mg/L, r=0.795, P=0.000] and DAS28 score [(4.20±0.73) points, r=0.703, P=0.000]. PTPN14 was positively correlated with TNF-α (r=0.633, P=0.000) and CRP level (r=0.435, P=0.013), but not with DAS28 score (r=0.033, P=0.856).ConclusionPTPs were expressed differently in peripheral mononuclear cells of RA patients. Among them, PTPN11 expression was correlated with inflammatory factors, CRP level and DAS28, which may serve as novel markers in RA diagnosis.
