不同浓度千金子对G250基因高表达肾癌Renca细胞株Livin蛋白表达的影响研究 被引量:2

摘要 目的观察不同浓度千金子对G250基因高表达肾癌Renca细胞株Livin蛋白表达的影响.方法将125只大鼠分为正常对照组、模型对照组、高剂量千金子治疗组、中剂量千金子治疗组以及低剂量千金子治疗组,每组大鼠25只.正常对照组正常饲养,其余各组采用皮下注射法构建肾脏移植瘤模型.高剂量千金子治疗组、中剂量千金子治疗组以及低剂量千金子治疗组分别于成瘤5d后连续给予千金子治疗,注射浓度分别为20mg/mL、10mg/mL以及2.5mg/mL,剂量为200mg/kg,1次/d,连续15d接受千金子治疗.正常对照组与模型对照组造模后给予注射等剂量生理盐水.比较各组大鼠抑瘤率、G250基因蛋白、Livin基因蛋白的表达.结果与模型对照组相比,高、中、低剂量千金子治疗组5-25d平均抑瘤率较高(P〈0.05),且各组平均抑瘤率呈逐渐增高趋势;与中剂量千金子治疗组以及低剂量千金子治疗组相比,高剂量千金子治疗组平均抑瘤率较高(P〈0.05).与正常对照组相比,模型对照组,高、中、低剂量千金子治疗组肾脏组织中Livin基因表达水平较低(P〈0.05);G250基因表达水平较高(P〈0.05);与中剂量千金子治疗组以及低剂量千金子治疗组相比,高剂量千金子治疗组Livin基因表达水平较高(P〈0.05);G250基因表达水平较低(P〈0.05).结论高浓度千金子对肾癌Renca细胞株具有良好的抑制作用,可能与下调G250基因表达,上调Livin蛋白表达相关. g Objective To observe the expression of Livin protein in high expression of G250 gene in renal cell carcinoma Renca cell line of different concentrations of moleplant seed. Methods One hundred twenty-five rats were divided into normal control group, model control group, high dose of moleplant seed group, middle dose of moleplant seed group and low dose of moleplant seed group, with 25 rats in each group. Normal control group was normal feeded, and the other groups were given subcutaneous injection to construct the kidney transplantation tumor model. High, middle and low dose of moleplant seed group were given 5 days consecutive moleplant seed treatment after the formation of tumor, injection concentration respectively 20 mg/mL and 10 mg/mL and 2.5 mg/mL dose of 200 mg/kg, 1 times per day, for 15 days accept moleplant seed treatment. Normal control group and model group were injected with normal saline after the models were made. The tumor inhibition rate, G250 gene protein and Livin gene protein of rats in each group were compared. Results Compared with model control group, the 5 to 25 days average inhibitory rate of high, middle and low dosage of moleplant seed treatment group was higher (P〈0.05), and the inhibitory rate was gradually increased. Compared with the middle and low dose moleplant seed treatment group, the inhibitory rate of high doses of moleplant seed treatment group average was higher (P〈0.05). Compared with the normal control group, the kidney tissue Livin gene expression level of model control group, high, middle and low dosage of moleplant seed treatment groups was lower (P〈0.05), and G250 gene expression levels were higher (P〈0.05). Compared with the middle and low dose moleplant seed treatment group, the Livin gene expression level of high dose of moleplant seed group were higher (P〈0.05), and G250 gene expression level was lower (P〈0.05). Conclusion High concentration of moleplant seed has a good inhibitory effect on renal cell carcinoma Renca cell line, which may be related to down regulation of G250 gene expression and up regulation of Livin gene protein expression.
