三黄益肾胶囊对链脲佐菌素诱导糖尿病大鼠心肌和肾组织结构的影响

Effect of Sanhuang Yishen Capsule on streptozotocin induced myocardial and renal tissue structure in diabetic rats

目的观察三黄益肾胶囊对链脲佐菌素诱导糖尿病大鼠心肌和肾组织结构的影响。方法采用链脲佐菌素腹腔注射制作糖尿病大鼠模型。造模后分组灌胃给药8周,检测各组大鼠空腹血糖(FPG),观察光镜下左室心尖部心肌组织及肾组织病理学改变。结果经三黄益肾胶囊干预8周后,与正常组比较,模型组大鼠心肌细胞肥大,心肌纤维排列紊乱,细胞间隙增大,胞浆染色不均匀,排列不规则,间质和血管外基质增多,并有炎症细胞浸润;三黄益肾胶囊中、高剂量治疗组大鼠介于正常组与模型组之间。模型组大鼠肾组织肾小球内皮细胞肿胀、系膜细胞增多;肾小管上皮肿胀,部分坏死;肾间质有炎性细胞浸润,血管扩张充血;三黄益肾胶囊中、高剂量治疗组大鼠介于正常组与模型组之间。低剂量治疗组较模型组心肌及肾组织均改善不明显。结论三黄益肾胶囊对链脲佐菌素诱导的糖尿病大鼠心肌细胞和肾组织有保护作用。

Objective It is to observe the effect of Sanhuang Yishen Capsule on myocardial and renal tissue of diabetic rats induced by streptozotocin（ STZ）. Methods Diabetic rat model was induced by intraperitoneal injection of streptozotocin. After the models were established, the rats were treated for 8 weeks by gavage, then the fasting blood glucose （FPG） was detected, and the pathological changes of cardiac tissue and renal tissue were observed under light microscope. Results After interven- tion of Sanhuang Yishen Capsule for 8 weeks, compared with normal group, myocardial hypertrophy, myocardial fibers ar- ranged in disorder,intercellular space increased, cytoplasmic staining uneven, irregular arrangement, an increase in interstitial and vascular extraeellular matrix and inflammatory cell infiltration were found in the rats in model group; the pathological chan- ges in medium and high dose of Sanhuang Yishen Capsule groups were between normal group and model group. Swelling of glo- merular endothelial cells and mesangial cells, renal tubule epithelial swelling and necrosis, kidney matter infiltration of inflam- matory cells, vascular dilatation and congestion were found in kidney tissue of the rats in model group, the pathological chan- ges in medium and high dose of Sanhuang Yishen Capsule groups were between normal group and model group. The myocardial and renal tissue were not significantly improved in low dose of Sanhuang Yishen Capsule group compared with that in model group. Conclusion Sanhuang Yishen Capsule has protecting effect on myocardial cells and renal tissue in diabetic rats induced by STZ.
