摘要
目的:研究大黄配伍药对后大黄中的大黄素、芦荟大黄素、大黄酸、大黄素甲醚、大黄酚、番泻苷A及番泻苷B的含量变化。方法:色谱柱为Dikma Dimonsil C_(18)(250 mm×4.6 mm,5μm),流动相为0.1%磷酸水溶液-乙腈,梯度洗脱,流速为1 mL·min^(-1),检测波长256 nm。结果:大黄及大黄药对的7个蒽醌类化合物的7条标准曲线在检测范围内均呈良好线性(r≥0.999);大黄及大黄药对的精密度RSD均小于3.0%;加样回收率为94.54%~103.1%。大黄各药对配伍后7个蒽醌类化合物含量均有不同程度的改变,与单味大黄比较,大黄桃仁、大黄牡丹及大黄甘遂组的番泻苷B、大黄酸、大黄素甲醚含量均升高,大黄枳实、大黄黄连组的番泻苷A、大黄素的含量均降低,其中降幅最大的是大黄黄连组,且大黄黄连组的大黄酚含量显著降低。结论:大黄配伍药对后会引起蒽醌类化合物含量的变化,其机制可能与配伍引起的大黄蒽醌类化合物溶出量变化及药对的相使相须作用有关,且该变化在一定程度上与大黄的功效存在共性关系。
Objective: To study on the changes of the 7 anthraquinione derivatives in the herb-pairof Radix et Rhizoma, including emodin, rhein, chrysophanol, physcion, aloe-emodin, sennoside A, and sennoside B. Methods : The analysis was achieved with a Dikma Dimonsil-C18 column (250 mm×4.6 mm,5 μm) , with gradi- entelution of 0. 1% (v/v) phosphoric acid in water and acetonitrile as the mobile phase. The flow rate was 1 mL·min^-1, and the detect wavelength was set at 256 nm. Results: All the calibration curves showed good linearity (r≥0. 999) in eetain concentration ranges. The RSD of the 7 analytes was all less than 3.0% , and the recovery rates were 94. 54% - 103. 1%. The 7 anthraquinione derivatives were changed after compatibility. Comparing with single Radix et Rhizoma, Radix et Rhizoma and Persieae Semem, the sennoside B, rhein and physcion in Radix et Rhizoma and Moutan Cortex, Radix et Rhizoma and Kansui Radix were all increased, the sennoside A and emodin in Radix et Rhizoma and Coptidis Rhizoma, Radix et Rhizoma and Aurantii Fructus Immaturus were decreased, especially, the group of Radix et Rhizoma and Coptidis Rhizoma. The ehrysophanol in Radix et Rhizoma and Coptidis Rhizoma was decreased significantly and sharply. Conclusion: The contens of anthraquinones and sennosides in Radix et Rhizoma were mostly changed by different compatibilities, which indicated that the mechanism in compatible herbs of Radix et Rhizoma affected the dissolution of anthraquinones, and the effect of the phase to the phase, which may contribute to the characteristics of efficacy of Radix et Rhizoma.
