摘要
采用响应面法优化啤酒糟中多糖的提取工艺,并对多糖的抗肿瘤活性进行评价。在单因素实验基础上,以浸提温度、浸提时间和固液比为影响因素,利用Box-Behnken中心组合方法进行三因素三水平实验设计,以多糖得率为响应值,进行响应面分析,采用MTT法分别考察啤酒糟多糖于24、48、72 h对He La、A549和Hep G2细胞增殖的抑制能力。结果显示,多糖提取的最佳工艺:浸提温度为96.7℃、浸提时间为1.83 h、固液比为1∶27 g/m L时,最大多糖得率为6.24%,验证值为6.07%,相对误差为2.72%;啤酒糟多糖对He La,A549和Hep G2细胞具有较好的抑制增殖能力,其72 h的IC50值分别为34.85、91.50和162.39μg/m L。表明本研究所得的啤酒糟多糖提取工艺高效、简单且可重复性强,同时该多糖可以作为一种潜在的辅助抗肿瘤成分应用在保健食品及医药领域。
Box-Behnken response surface method was to optimize extraction process of polysaccharide from beer vinasse and the antitumor activity of polysaccharide was evaluated.Based on the single factor experiment, leaching temperature, leaching time and solid liquid ratio as factor, the Box-Behnken central composite method was used to make three factors, three levels experimental design.The polysaccharide yield was the response value to process response surface analysis.Adopting MTT method was inspected the inhibition capability of polysaceharide from beer vinasse at 24,48,72 h to the cell proliferation of HeLa, A549 and HepG2.It turned out that the best process of polysaccharide was when leaching temperature was 96.7 ℃, leaching time was 1.83 h, solid liquid ratio was 1:27 g/mL, the maximum polysaccharide yield was 6.24% ,verifying value was 6.07% , relative error was 2.72%.Polysaccharide of beer vinasse could better inhibit multiplication capacity was 34.85,91.50 and 162.39 μG/mL. It showed that the extraction process of polysaccharide from beer vinasse in the study was effective, easy and repeatable.Meanwhile, the polysaccharide could be a potential auxiliary antitumor components to apply in health food and medicine field.
