盐敏感性高血压大鼠外周血液白细胞端粒长度的变化

Changes of peripheral leukocytes telomere length in salt-sensitive hypertension rats

摘要 【目的】探讨高血压大鼠外周血液白细胞端粒长度的变化。【方法】雌性SD大鼠30只,随机分为两组:高血压组和假手术组,每组15只,采用双侧肾动脉后支结扎、术后予以含8%Na Cl饲料构建高血压大鼠模型。选取术后4个月为观察终点,此时行眶下静脉丛取血,提取血液基因组DNA并应用real-time PCR法测量端粒相对长度,分离血浆并应用ELISA法测量血浆端粒酶活性。【结果】与对照组比较,高血压组大鼠端粒长度明显缩短(0.83±0.23 vs.1.04±0.29,P<0.05),血浆端粒酶活性变化无显著差异(74.43±13.04 vs.69.71±11.11,P>0.05)【结论】在大鼠体内,端粒长度缩短与高血压疾病密切相关,但相关机制可能与端粒酶修复失衡无关,端粒在高血压的发展中可能发挥着重要作用。 [Objective ] To explore the changes of telomere length in the peripheral leukocytes of hypertensive rats. [ Methods] A total of 30 female SD rats were randomly divided into 2 groups equally: hypertensive group and sham group. Rats in the hypertensive group were operated by ligating the posterior branches of bilateral renal arteries, and fed with high salt diet (8% NaC1). When the operation was performed after 4 months, genomic DNA was extracted from the blood of inferior orbital venous plexus. Telomere length was measured by real-time PCR, and telomerase activity in plasma was determined by ELISA. [ Results] Compared with the sham group, the telomere length in the hypertensive group was significantly shortened (0.83 ± 0.23 vs. 1.04 ± 0.29, P〈0.05), while the telomerase activity showed no significant difference (74.43 ± 13.04 vs. 69.71 ± 11.11, P〉0.05). [Conclusion] Telomere shortening is closely related with hypertension in rats, but the mechanism may not relate to the imbalance of telomerase repair. Telomere may play an important role in the development of hypertension.
