MitoQ激活Nrf2抗氧化系统缓解高脂诱导的胰岛素抵抗

摘要 目的观察米托蒽醌甲磺酸盐(mitoquinone mesylate,MitoQ)改善高脂肪所致的胰岛素抵抗作用并探讨其改善胰岛素抵抗的效应机理。方法将30只C57BL/6J雄性小鼠随机分成低脂组(LFD组)10只、高脂组(HFD组)20只,分别给予低脂饲料、高脂饲料喂养12周后,采用葡萄糖耐量试验(IPGTT)检测胰岛素抵抗模型,模型成功后,HFD组随机选取10只进行米托蒽醌甲磺酸盐灌胃干预(MitoQ组)。30 d后取胰岛素敏感组织肌肉,用免疫印迹方法(Western blot,WB)检测蛋白水平和磷酸化变化。结果在HFD诱导的小鼠胰岛素抵抗模型中,MitoQ组可减少小鼠葡萄糖耐量并纠正高脂所致的胰岛素信号蛋白PKBSer473磷酸化损害;MitoQ组改善HFD饲喂小鼠损害的Nrf2系统功能,促进Nrf2核转位,同时明显降低Nrf2抑制性蛋白Keap1的表达。结论线粒体抗氧化剂MitoQ通过增强肌肉Nrf2系统功能和胰岛素信号水平,缓解高脂肪诱导的胰岛素抵抗。
