摘要
[目的]观察和分析中药疏肝健脾方对腹泻型肠易激综合征(IBS-D)模型大鼠结肠神经生长因子(NGF)及其受体TrkA、P75表达的影响,以期阐述中药疏肝健脾方治疗IBS-D可能机制。[方法]将18只2日龄SD雄性幼鼠随机分为正常组、模型组、疏肝健脾组,除正常组外其余各组使用母婴分离加束缚应激的方法制备IBS-D大鼠模型。造模成功后正常组、模型组予生理盐水,疏肝健脾组予疏肝健脾方灌胃14d。取大鼠结肠黏膜,采用酶联免疫法检测NGF及其TrkA、P75表达。[结果]疏肝健脾方可以降低IBS-D模型大鼠结肠黏膜组织NGF含量(P>0.05)、5-HT含量(P<0.05),下调受体TrkA、P75的表达(P<0.05)。[结论]疏肝健脾方干预IBS-D模型大鼠,改善内脏敏感性及腹泻部分机制与调节NGF及其TrkA、P75表达有关。
[Objective]To investigate and analysis the effects of Shuganjianpi preseription(SGJP)on the expression of nerve growth factor(NGF), Trk A receptor and P75 receptor of colon mucosa in the irritable bowel syndrome-diarrhea predominant (IBS-D)model rats. [Methods] Eighteen 2-day-old male Sprague- Dawley(SD) rats were randomly divided into three groups:normal group, model group, and SGJP group. All groups except the normal group received neonatal maternal separation plus restraint stress for preparation of IBS-D model,which were intragastric administrated with saline and SGJP for 14 days respectively after modeling successfully. The colon mucosa of rats was taken for detecting the expression of NGF, Trk A re- ceptor and P75 receptor by enzyme linked immunosorbent assay. [Results]The SGJP reduced the expression of NGF(P〉0.05),TrkA receptor(P〈0.05)and P75 (P〈0.05)receptor of colon mucosa in IBS-D rats. [Conclusion]The partial mechanisms of improving the visceral sensation and diarrhea with SGJP in IBS-D rats might be realized through regulating NGF and its receptors.
