摘要
目的探讨维生素E对体外人绒毛膜滋养层细胞的保护作用及机制,为维生素E保胎机制提供理论基础。方法以体外培养的滋养层细胞为模型,采用实时荧光定量PCR的方法检测维生素E对体外培养的滋养层细胞中Foxo3a和Survivin表达水平的影响,采用酶联免疫吸附方法分析维生素E对体外培养的滋养层细胞中半胱氨酸天冬氨酸蛋白酶-3(Caspase3)活性的影响。结果维生素E下调Foxo3a表达水平;上调凋亡抑制基因Survivin表达水平;下调Caspase3活性水平,且250 mg/L实验组结果差异最显著。结论维生素E可能通过抑制Foxo3a的表达,从而促进Survivin表达,降低Caspase3活性水平,达到抑制细胞凋亡的作用,此研究为临床应用维生素E保胎治疗提供理论基础,为提高生育率、减少自然流产率及提高人口素质打下坚实的基础,具有重要的临床及社会意义。
Objective To explore the protective effect and mechanism of vitamin E on human trophoblast cells in vitro, and provide a theoretical basis for vitamin E in tocolytic therapy. Methods Taking human trophoblast cells cultured in vitro as model, real-time fluorescent quantitative PCR was used to detect the effect of vitamin E on the expression levels of Foxo3a and Survivin, ELISA was used to analyze the effect of vitamin E on Caspase-3 activity. Results The expression rate of Foxo3a was down-regulated by vitamin E, the expression rate of Survivin was up-regulated, and Caspase-3 activity was down-regulated, the difference of results in 250 mg/L experimental group was the most significant. Conclusion Vitamin E may decrease the activity of Caspase-3 to inhibit apoptosis by inhibiting the expression of Foxo3a and promoting the expression of Survivin. The study provides a theoretical basis for clinical application of vitamin E in tocolytic therapy and lays a solid foundation for the improvement of fertility, reduction of spontaneous abortion rate, and the enhancement of population quality, which has important clinical and social significance.
