超声-微波协同提取昆仑雪菊色素工艺及其抗氧化活性研究 被引量:4

Optimization of ultrasonic-microwave synergistic extraction of pigment from Kunlun Coreopsis tinctoria and its antioxidant activity

摘要 以新疆昆仑雪菊为原料,采用超声-微波协同提取昆仑雪菊色素,在单因素试验的基础上,采用响应面试验设计,以雪菊色素色差值(E)为响应值,以浸取温度、浸提时间和料液比为影响提取过程中的3个因素进行试验设计;并测定雪菊色素提取液对DPPH自由基(DPPH·)、羟自由基(·OH)和超氧阴离子自由基(O^(2-)·)的清除能力。结果表明:超声-微波协同提取昆仑雪菊色素最佳工艺条件为:提取时间20 min,提取温度44℃,料液比1∶21,乙醇浓度50%,微波功率120 W,在此条件下提取的雪菊色素E为54.72,经过实际试验重复验证,平均雪菊色素E为53.7,相对误差为3.14%与预测值比较接近;雪菊色素对DPPH·、·OH和O^(2-)·清除作用明显,表明雪菊色素具有良好的抗氧化作用,可以作为天然抗氧化剂和进一步开发和利用。 Based on the single factor experiment, pigment from Coreopsist tinctoria groun in Kunlun, Xinjiang was extracted by ultrasonic-microwave synergistic method. The experiment was designed by response surface method and E value of snow chrysanthemum was selected as the response value. The extraction temperature, extraction time and solid-liquid ratio were 3 factors which influenced extraction process. The scavenging capacity of Coreopsis tinctoria pigment solution was measured by DPPH, hydroxyl radical and superoxide radical. The results showed that the best conditions for extraction of pigment were : extraction time 20min, extraction temperature 44℃, solid-liquid ratio 1 : 21, ethanol concentration 50%, microwave power 120W. The E value of the extracted pigment was 54.72 under this condition. After the repeated verification from actual test, the average value of E was 53.7, the relative error was 3.14% and the predicted value was close to the average. The pigment from Coreopsis tinctoria showed significant scavenging effects on DPPH · , · OH and O^2- . The pigment from Coreopsis tinctoria with excellent antioxidant has the potential to be further developed and utilized as a natural antioxidant.
