HIV/AIDS肺脾气虚证与气阴两虚证患者尿液的代谢组学研究 被引量:10

Metabonomics Study on the Urine of Patients with HIV/AIDS Syndrome of Qi Deficiency of Lung and Spleen and Deficiency of Both Qi and Yin

摘要 目的探讨代谢组学技术在中医证型诊断中的价值。方法 2011年12月选取河南省某地区人类免疫缺陷病毒(HIV)/艾滋病(AIDS)患者23例,其中脾肺气虚证患者12例作为HIV/AIDS肺脾气虚证组,气阴两虚证患者11例作为HIV/AIDS气阴两虚证组。运用液相色谱质谱分析(LC-MS)检测23例HIV/AIDS患者的尿液代谢成分,结合主成分分析(PCA)和t检验分析技术,分析HIV/AIDS患者尿液的差异代谢物。结果通过t检验结合1.5倍原则对数据进行的初步筛选,发现质核比(m/z)为317、213的可能存在的代谢物为两组的差异代谢物,其峰强度在HIV/AIDS气阴两虚证组患者中均低于HIV/AIDS肺脾气虚证组患者(P<0.05)。PCA显示m/z为967、155、317、213的离子物与其他离子物相距较远,可以认为是HIV/AIDS肺脾气虚证组和HIV/AIDS气阴两虚证组间潜在的差异代谢物。结论运用代谢组学技术中的LC-MS可以将HIV/AIDS肺脾气虚证和HIV/AIDS气阴两虚证尿液在代谢物轮廓上进行区分,实现不同中医证型间的差异代谢物比较,代谢组学技术在指导中医证型的诊断方面有良好的应用价值。 Objective To investigate the value of metabonomics technology in the diagnosis of TCM syndrome types.Methods A total of 23 patients with human immunodeficiency virus(HIV)/acquired immune deficiency syndrome(AIDS) in an area of Henan Province in December 2011 were selected.Among them,12 with syndrome of Qi deficiency of lung and spleen were taken as HIV/AIDS group of syndrome of Qi deficiency of lung and spleen,and 11 with syndrome of deficiency of both Qi and Yin were taken as HIV/AIDS group of syndrome of deficiency of both Qi and Yin.The metabolic components of urine of 23 patients with HIV/AIDS were detected by liquid chromatography-mass spectrometry(LC-MS),and the differential metabolites of urine of patients with HIV/AIDS were analyzed by combining with analysis techniques of principal component analysis(PCA) and t test.Results Through the t test and principle of 1.5 times,the preliminary screening of data found that the possible metabolites with karyoplasmic ratio(m/z) of 317 and 213 were differential metabolites of two groups,the expression changes of the peak intensity in HIV/AIDS group of syndrome of deficiency of both Qi and Yin were lower than those in HIV/AIDS group of syndrome of Qi deficiency of lung and spleen(P〈0.05).PCA showed that ions with m/z of 967,155,317 and 213 ions were far away from other ions,which could be considered as the potential differential metabolites between the two groups.Conclusion The LC-MS in metabonomics technologies can differentiate the urine of patients between the two groups from the profile of metabolites,and achieve the comparison of differential metabolites in different TCM syndromes.The metabonomics technologies has a good application value in guiding diagnosis of syndrome of TCM.
