摘要
目的探讨对急性肝衰竭大鼠模型采用大黄提取物的治疗作用。方法利用D-氨基半乳糖建立大鼠急性肝功能衰竭模型。选取SD大鼠15只,随机分为3组(n=5)。A组为健康大鼠,不施加任何试验处理;B组为肝衰竭大鼠,造模后12 h尾静脉注射1 mL生理盐水;C组为大黄提取物治疗组,造模后12 h向腹腔注射大黄提取物(10 mL/kg),每日3次,连续注射2 d。各组大鼠均在造模48 h之后注射10%的水合氯醛进行麻醉,取门静脉血以及肝组织标本进行试验指标观测。结果 B组大鼠造模后出现急性肝衰竭表现,而A组、C组均无或仅有少量症状出现。B组大鼠在造模48 h后血清ALT、AST以及TBiL含量均出现显著上升,而C组出现下降。结论大黄提取物有利于促进急性肝衰竭大鼠的恢复,并且能促进骨髓间充质干细胞的增殖,增强BMSCs细胞移植的治疗效果。此治疗方法能够促进肝细胞的再生和修复,为肝功能障碍患者提供一定的治疗。
Objective To explore the therapeutic effect of rhubarb extract in the model of rats with acute liver failure. Methods D-galactosamine was used to establish the model of rats with acute liver failure. A total of 15 SD rats were selected and randomly divided into 3 groups （n=5）. Group A was healthy rats without any experimental treatment; group B was ＂the rats with liver failure, and the rats were injected with 1 mL normal saline at caudal vein 12 hours after modeling; group C was the treatment group of rhubarb extract, at 12 hours after modeling at caudal vein, rhubarb extract （10 mL/kg） was intraperitoneally injected 3 times a day for a continuous injection of 2 days. Rats in each group were injected with 10% of chloral hydrate after anesthesia for 48 hours. Portal vein blood samples and liver tissue specimens were collected for the observation of test indices. Results Rats in group B showed acute liver failure after modeling, and group A, group C were without or only with a small number of symptoms. The levels of serum ALT, AST and TBiL in group B 48 h after modeling were significantly increased, and the levels were reduced in group C. Conclusion Rhubarb extract is beneficial to promoting the recovery of acute liver failure rats, and can promote the proliferation of bone marrow mesenchymal stem cells and enhance the therapeutic effect of BMSCs transplantation. This therapy can promote the regeneration and repair of liver cells, and provide certain treatment for the patients with liver dysfunction.
