摘要
目的对纯钛表面进行超声微弧氧化(MAO)-多巴胺载葛根素处理,研究经不同浓度葛根素处理试件的抗菌性及对成骨细胞生物相容性的影响。方法以纯钛MAO(微弧氧化)浸多巴胺24 h后的试件为A组,纯钛MAO(微弧氧化)浸多巴胺24 h后分别载0.15、0.30、0.45 g/20mL的葛根素处理的试件为B、C、D组。将四组试件分别与大肠杆菌和金黄色葡萄球菌共同培养,通过全自动菌落计数仪计算其抗菌性。通过碱性磷酸酶(ALP)和Cell Counting Kit(CCK-8)试剂盒、扫描电镜、激光共聚焦显微镜等方式检测不同处理膜层的细胞相容性。结果 C、D两组涂层对金黄色葡萄球菌和大肠杆菌具有较好的抗菌性能,当葛根素的添加量达0.30 g/20 mL时,抗菌效果可达99%以上。ALP和CCK-8检测结果显示,四组材料表面细胞的增殖、活性顺序为C优于D优于B优于A组,其中C组具有最优的成骨细胞相容性。据统计学分析,A、B、C、D四组结果均具备统计学意义。结论纯钛MAO(微弧氧化)浸多巴胺载葛根素处理后涂层可增强试件的抑菌性和细胞相容性。当葛根素的浓度达0.30 g/20 mL时,涂层具有最优的抑菌性和生物相容性。
Abstract : Objective Study the effects of different concentrations of puerarin on the antimicrobial prop-erties of the specimens and osteoblast biocompatibility by processing pure titanium surfaces with ultrasonic micro - arc oxidation (MAO) - dopamine puerarin treatment . Method The specimens of group A were pure titanium treated with MAO leaching dopamine for 24 h. , The specimens of groups B, C, D were treated with MAO leaching dopamine for 24 h plus 0. 15, 0. 15, 0.45 g/20 L of puerarin respectively. The four groups of specimens were incubated with Escherichia coli and Staphylococcus aureus. Their anti- microbial activitis were calculated by automatic colony counting. Cell compatibility of different membranes was measured by alkaline phosphatase (ALP) and Cell Counting Kit (CCK 8) Kit, scanning electron microscopy (sem), laser confocal microscope . Results Groups C and D has good antibacterial properties against staphylococcus aureus and escherichia coli. When the amount of puerarin was 0. 30 g/mL, the anti- bacterial effects was over 99%. ALP and CCK 8 tests shows that, for the four groups of material surface, the cell value enhancement and activity is of the order that C is better than D better than B better than A. Group C has the best osteoblast compatibility. According to statistical analysis, results from A, B, C, D four groups were statistical significant. Conclusion The pure titanium MAO leaching dopamine plus puerarin treatment of the coating can enhance the antibacterial property and cell compatibility of the specimen.
