摘要 目的:研究苦远志中糖酯类化学成分及其抗氧化及抗黄嘌呤氧化酶活性。方法:采用D101大孔树脂、硅胶柱层析、Sephaclex LH-20、半制备高效液相色谱等多种方法分离纯化,并结合MS、NMR等波谱分析方法鉴定化合物的结构。利用ABTS方法和尿酸生成法分别测定其抗氧化和抗黄嘌呤氧化酶的活性。结果:从苦远志乙醇提取物中分离得到8个糖脂类化合物,分别鉴定为:蔗糖-6-苯甲酸酯(1)、sibiricose A6(2)、sibiricose A1(3)、glomeratose B(4)、sibiricose A3(5)、glomeratose A(6)、glomeratose D(7)、tenuifoliside C(8)。化合物4、7和8表现出较明显的抗氧化活性和抗黄嘌呤氧化酶抑制活性。结论:所有化学成分均为首次从该植物中分离得到,糖酯类成分在抗氧化与抗黄嘌呤氧化酶趋势上基本一致。 Objective:To study the inhibitory activity against xanthine oxidase and the antioxidant activity of glycolipids from Polygala sibirica var.megalopha.Methods:The products were isolated and purified by resin D101,silica gel,Sephadex LH-20 column chromato-graphy and preparative HPLC.Their structures were elucidated on the basis of various spectroscopic analysis,the antioxidant and anti-xanthine oxidase activities were tested by ABTS and uric acid generation methods.Results:Eight compounds were isolated from the ethanol extract of Polygala sibirica var.megalopha and identified as 6-O-benzoyl sucrose(1),sibiricose A6(2),sibiricose A1(3),glomeratose B(4),sibiricose A3(5),glomeratose A(6),glomeratose D(7),tenuifoliside C(8).Compound 4,7,8 showed strong antioxidant and anti-xanthine oxidase activities.Conclusion:Eight glycosides are isolated from this plant for the first time,the inhibitory activity against xanthine oxidase and the antioxidant activity of glycolipids trends are equally.
