具有miRNA海绵功能的circRNA的生物信息学分析与鉴定 被引量:1

摘要 目的对一种新的具有miRNA海绵功能的circRNA_hsa_circ_0000254结构、功能进行生物信息学分析与鉴定。方法采用生物信息学对hsa_circ_0000254的二级结构及靶向序列进行分析,通过双荧光素酶实验验证hsa_circ_0000254与靶基因的结合情况。结果生物信息学分析发现hsa_circ_0000254,剪接序列长度为524nt,包含11个miR_125b可结合位点;双荧光素酶实验证实hsLcirc_0000254与miR_125b具有较高的结合效率。,结论hsa_circ_0000254能以miRNA海绵形式抑制miR-125b,有可能作为靶向治疗工具应用于高表达miR-125b的白血病治疗。 Objective The bioinformatics analysis and identification of structure and function of a new circRNA- hsa circ 0000254 with miRNA sponge fimction. Methods The secondary structure and the targeting sequence of hsa circ 0000254 was identified by bioinformatics analysis. Dual-luciferase reporter assay system was used to determine the influence of hsa_circ_0000254 on its target gene. Results The bioinformatics analysis revealed that the splicing sequence of hsa-circ-000254 was 524nt and contained 11 binding sites of miR-125. Dual-luciferase reporter assay system revealed that miR-125b could significantly diminish luciferase activity by the reporter vector contained the 3' UTR of hsa-circ-000254. Conclusion The results strongly suggest that hsa- circ-000254 can effectively combine with miR- 125b and may be acted as a new molecular target therapy for leukemia which highly expressed miR-125b.
