壮医方白花丹消蛊汤对肝纤维化食蟹猴的干预作用及其机制 被引量:4

摘要 目的探讨壮医方白花丹消蛊汤对肝纤维化食蟹猴的干预作用及其机制。方法 12只食蟹猴随机分为空白组、模型组、白花丹消盅汤低剂量组、高剂量组,每组3只。采用四氯化碳复合高脂饮食诱导法建立食蟹猴肝纤维化模型,造模成功后空白组及模型组给予生理盐水灌胃,白花丹消蛊汤低剂量、高剂量组分别给予1.0 g/(kg·d)、4.0 g/(kg·d)的汤剂。8周后检测各组食蟹猴外周血ALT、AST、AKP、TBIL、Ⅰ型胶原(COL-Ⅰ)、Ⅲ型胶原(COL-Ⅲ)及Ⅳ型胶原(COL-Ⅳ)水平,肝穿获取肝组织标本,行天狼猩红染色后观察肝组织胶原变化。结果与模型组相比,白花丹消蛊汤高剂量组食蟹猴血清ALT、AKP、TBIL水平更低,且血清COL-Ⅰ、COL-Ⅲ及COL-Ⅳ含量更低,肝纤维化胶原异常增生程度得到改善。结论壮医方白花丹消蛊汤具有抗肝纤维化作用,可能是通过减少胶原异常增生发挥作用。 Objective To explore the intervention effect of Zhuang medicine prescription of Baihuadan Xiaogu Tang on cynonlolg-s macaque with hepatofibrosis and its mechanism. Methods Twelve cynomolgus macaques were randomly divided into blank group, model group,low-dose Baihuadan Xiaogu Tang group and high-dose Baihuadan Xiaogu Tang group ,with 3 macaques in each group. The cynomolgus macaque model with hepatofibrosis was established using carbon tetractdoride combined with fat-rich diet. After successful modeling,intragastric -dministration of normal saline was performed in the blank group and model group. Low-dose Baihuadan Xiaogu Tang group and high-dose Baihuadan Xiaogu Tang group were administered the decoction with the dose of 1.0g/( kg - d) and 4.0g/( kg - d) respectively. Eight weeks later,the levels of peripheral ALT, AST, AKP, TBIL, collegen (COL) - [ , COL-llI and COL-IV were detected in the cynomolgus macaques of each group. The liver tissue specimens were obtained by liver biopsy and stained by sirius red for an observatiou on the change of COL in the liver tissues. Results Compared to the model group,the levels of serum ALT, AKP and TBIL as well as the levels of COL-Ⅰ, COL-Ⅲ and COL-Ⅳwere lower in the cynomolgus macaques of high-dose Baihuadan Xiaogu Tang group ,and improved COL proliferation abnormality was achieved in eynomolgus macaques with hepatofibrosis. Conclusion Zhuang medicine prescription of Baihuadan Xiaogu Tang obtains anti-hepatofibrosis effect probably through decreasing COL proliferation abnormality.
