清眩降压汤控制自发性高血压大鼠血压升高与抑制心肾大血管损害的药效学研究

Pharmacodynamics of Qingxuan Jiangya Decoction on Blood Pressure Control and Suppression of Cardiac and Renal Vascular Injury in Spontaneously Hypertensive Rats

目的:探讨清眩降压汤对自发性高血压大鼠血压控制和心肾血管的影响。方法:将12只自发性高血压大鼠随机分为2组,每组各6只。中药组给予清眩降压汤[5 g/(kg·d)]灌胃,模型组给予等容积生理盐水灌胃,连续干预8周。另取6只Wistar大鼠作为正常血压对照组(正常组),给予等容积生理盐水灌胃。给药期间,每周测量血压和体质量。8周后,麻醉取胸主动脉、心脏、肾脏进行HE染色,测量血管中膜厚度(MT)和管腔直径(LD),计算LD/MT比值。结果:清眩降压汤干预3周后,中药组大鼠的收缩压、舒张压及平均动脉压均低于模型组(P<0.05);中药组血管中膜厚度较模型组薄,LD/MT比值小(P<0.05);与模型组比较,中药组大鼠肾小球固缩,心肌纤维肿胀变性,横纹结构不清,排列紊乱显著改善。结论:清眩降压汤可控制自发性高血压大鼠血压的进一步升高,抑制血管重构进程,改善血压升高导致的心肾等靶器官的损害。

Objective：To investigate Qingxuan Jiangya decoction on modulation of blood pressure and protection of cardiac, renal and vascular from remodeling in spontaneously hypertensive rats. Methods：A total of twelve spontaneously hypertensive rats were randomly assigned into two groups. The treatment group was given Qingxuan Jiangya decoction [5 g（kg·d）], and the model group was given the same volume of saline, six Wistar rats were used as the normal group. The intervention process lasted for eight weeks. Blood pressure and body weight were measured weekly. Rats sacrificed at ending point, and the thoracic aorta, heart and kidney were then taken for HE staining. The thickness of vascular membrane（MT） and lumen diameter（LD） were measured, and the ratio of LD/MT was calculated. Results：The systolic blood pressure, diastolic pressure and mean of arterial pressure of rats in the treatment group were lower than those of model group（P〈0.05） after received Qingxuan Jiangya decoction for three weeks. Compared with the model group, the arterial intima-media of rats in treatment group became thinner, and the ratio of LD/MT became smaller（ P〈0.05）. The glomerular pyknosis, disorder of myocardial fiber arrangement in the rats of treatment group improved significantly compared with those of model group. Conclusion：Qingxuan Jiangya decoction can control further increasing of blood pressure, inhibit vascular remodeling process, protect target organs such as heart and kidney from the damage caused by high blood pressure.
