摘要
目的:探讨活血药川芎对高转移性人巨细胞肺癌细胞PG-BEI干细胞样细胞荷瘤裸鼠手术前后HIF-1α蛋白表达的影响。方法:分选PG-BG1中的干细胞样细胞亚群,并将其接种于裸鼠右下肢。根据干预时间和药物的不同,分为对照组、乏氧组、川芎术前组、川芎术后组、顺铂术前组、顺铂术后组、川芎联合顺铂术前组和川芎联合顺铂术后组。于第21天行截肢手术并对原位瘤组织称重,计算抑瘤率,用Western blot法检测HIF-1α表达。第40天获取小鼠复发瘤组织,并对复发瘤组织称重,计算复发率、复发率抑制率、复发瘤重及复发瘤重抑制率,Western blot法检测HIF-1α表达结果。结果:肺癌干细胞样细胞荷瘤裸鼠原位瘤方面,川芎联合顺铂组原位瘤重及HIF-1α表达较空白组和单纯顺铂组减少。复发瘤组织方面,与模对照、化疗组比较,川芎联合顺铂组复发率、复发率抑制率、复发瘤重、复发瘤重抑制率及HIF-1α表达明显下调。低氧环境较常氧环境下HIF-1α表达增加。结论:川芎联合顺铂组可以通过抑制HIF-1α的表达从而抑制肺癌干细胞样细胞荷瘤裸鼠肿瘤组织的增殖和复发。
Objective:To explore the effect of Rhizoma Chuanxiong (CX)on HIF - 1 α expression of tumor bearing nude mice before and after surgery in human lung cancer stem cell line PG. Methods : Select stem cell like cells in PG - BG1 and inoculate them in the right lower limb of nude mice. According to the period and the drug types of intervention, the nude mice were divided into eight groups : the blank control group, the hypoxic group, the pre - op CX group, the post - op CX group, the pre - op cisplatin group, the post - op cisplatin group , the pre - op CX plus cisplatin group and the post - op CX plus cisplatin group. On the twenty - first day ,perform an amputation. Weigh in situ tumor tissue. Calculate the tumor inhibition rate and detect the expression of HIF - 1 alpha by Western blot method. On the fortieth day, get recurrent tumor tissue in mice. Weigh in recurrent tumor tissue. Calculate the recurrence rate, inhibition rate of recurrence rate, recurrent tumor weight and recurrent tumor weight inhibition rate and detect the expression of HIF - 1 alpha by Western blot method. Results : In primary tumor of lung cancer stem cell like cells in nude mice, the situ tumor weight and HIF - 1 expression decreased in CX plus with cisplatin group compared with the blank group and the cisplatin group. In recurrent tumor tissue, the recurrence rate, inhibition rate of recurrence rate, recurrent tumor weight, recurrent tumor weight inhibition rate and HIF - 1 expression decreased in CX plus with cisplatin group compared with the blank group and the cisplatin group. The expression of HIF - 1 was increased in hypoxia environment than in normal oxygen environment. Conclusion:CX plus cisplatin group can suppress tumor tissue proliferation and recurrence of tumor bearing nude mice in lung cancer stem cell like cells through inhibiting the expression of HIF - 1α.
