摘要
目的:通过预实验观察化痰祛湿活血方对非酒精性脂肪性肝炎(NASH)大鼠血清及肝组织中相关氧化应激因子的影响,初步探讨其治疗NASH的作用机制。方法:SPF级SD雄性大鼠随机分为空白组、模型组、对照组、高剂量组、中剂量组、低剂量组,空白组予普通饲料,其余组给予高脂饲料联合四环素腹腔注射造模。造模第9周处死全部实验动物,HE染色观察肝组织病理结构,ELISA法测定血清及肝组织中MCP-1、PTGS2、NOS2、MDA含量。结果:各用药组血清及肝组织中MCP-1、PTGS、NOS2、MDA含量均低于模型组(P<0.01或P<0.05),高剂量组降低最显著,优于对照组(P<0.05)。结论:化痰祛湿活血方可减轻NASH大鼠炎症损伤,其机制可能与其能够下调氧化应激相关因子MCP-1、PTGS2、NOS2、MDA表达有关。
Objective : To observe the effects of Huatan Qushi Huoxue Formula on expression of oxidative stress related factors in the serum and hepatic tissue of NASH rats and investigate its mechanism through the pre - experiment. Methods:Totally 72 male SD rats were randomized into the blank group, model group, control group and Huatan Qushi Huoxue Formula high - dose, middle -dose and low -dose groups,with 12 in each. After the ninth week,the liver tissue sections were stained by HE staining and liver tissue homogenate. The expressions of MCP - 1, PTGS2, NOS2 and MDA were detected by enzyme linked immune sorbent assay (ELISA). Results:Compared with the model group,the treatment group can effectively reduce the MDA, NOS2,PTGS2 and MCP - 1 in the serum and hepatic tissue( P 〈 0. 01 or P 〈 0. 05 ). The effect of high dose group was significantly better than the control group' s ( P 〈 O. 05 ). Conclusion : Huatan Qushi Huoxue Formula could alleviate the inflammatory damage in NASH rats, the mechanism of which might be related to the down - regulation of MCP - 1, PTGS2, NOS2 and MDA.
