摘要 目的:对甜茶多酚的安全性进行评价。方法:将40只小鼠随机分为给药组与空白组,给药组每天给予小鼠灌胃0.4mL·20g^(-1) Body weight(BW)的甜茶多酚,空白组给予等量生理盐水,给药7天,观察小鼠的体重、进食量及组织病理学等变化以评价甜茶多酚的急性毒性。将120只大鼠分为甜茶多酚低、中、高剂量(27g·kg^(-1)·d^(-1)、54g·kg^(-1)·d^(-1)、108g·kg^(-1)·d^(-1))组及空白对照组,每天灌胃1次,空白对照组给予等量蒸馏水。观察各组大鼠的一般状况及给药12周、停药2周后大鼠的体重、脏器指数、血液学、血液生化学及病理组织学变化以评价甜茶多酚的长期毒性。结果:急性毒性:小鼠给予大剂量甜茶多酚,第3天后,小鼠的进食量、体重、肝病理切片与空白组比较无统计学差异(P>0.05);长期毒性:中剂量组、高剂量组大鼠的体重均明显降低(P<0.05);中剂量组大鼠的心脏指数明显增高(P<0.05),高剂量组大鼠的脾脏指数明显降低(P<0.05),停药恢复后心、肝、脾、肺、肾的脏器指数与空白组比较均无统计学差异(P>0.05);高剂量组大鼠Na^+浓度显著性增高外(P<0.05),血液生化学、血液电解质与空白组比较无统计学差异(P>0.05)。结论:甜茶多酚口服给药具有较好的安全性。 Objective: To study the acute toxicity in mice and long-term toxicity in rats to evaluate the safety of Rubus Suavissirnus. Methods:Acute toxicity observation:Forty mice were randomly divided into two groups, medication group were given intrgastric administration once a day, the control group were given equal-volume normal saline; Long-term toxicity observation : the total of 120 rats were randomly divided into the high, middle, and low doge groups. Which were treated with Rubus suavissirnus(27.54 and 108g·kg^-1·d^-1) by intragastric administration, once a day for 12 weeks. The general behaviors and physical status, hematology blood biochemistry, organs coefficient and histopathological analysis were detected in 12 weeks and 2weeks after drug. Results: Acute toxicity observation : Mice were given large doses of sweet tea polyphenols, paragraph 3d later, the body of mice food intake, body weight, slices of liver compared with the control group no significant difference. Long term toxicity observation: Middle dose group and high dose group body weight of rats were significantly decrease(P〈0.05). The cardiac index of middle group were significantly increased and the splenic index of high group were significantly decrease(P〈0.05) compared with the control group , while which were reversed to normal in 2 weeks after drug withdrawal. Except the sodium ion concentration of high group were significantly increased , Blood chemistry and blood electrolyte with the control group no significant difference. Conclusion: Rubus suavissirnus have higher security by oral.
