摘要 目的:建立HPLC-ELSD法测定黄芪注射液中单糖和蔗糖含量。方法:色谱柱为Alltech Prevail Carbonhydrate Polymeric NH_2(聚合物氨基柱)(250mm×4.6mm,5μm),以水-乙腈为流动相梯度洗脱,流速为1.0mL/min,检测器为蒸发光散射检测器,漂移管温度为80℃,气体流速为2.0L/min,柱温为35℃。结果:该条件下D-木糖、D-果糖、D-葡萄糖和蔗糖线性范围分别为0.26~2.60mg/min,0.28~2.80mg/min,0.14~1.40mg/min与1.40~14.00mg/min;平均回收率均在95.0%~105.0%范围内,RSD均<2.0%。结论:建立的方法可靠、简单、重复性好,可用于同时测定黄芪注射液中单糖和蔗糖含量,为黄芪注射液的质量控制提供参考。 Objective : To establish an HPLC-ELSD method for determination of monosaccharide and sucrose components simultaneously in Astragalus injection. Methods:Chromatographic separation was performed on an Alltech Prevail Carbonhydrate Polymeric NH2 column(250mm× 4.6mm, 5μm) using a mobile phase composed of a mixture of acetonitrile and water with gradient elution at a flow rate of 1.0mL/min. Detector was ELSD and column temperature was 35 ℃. The ELSD drift tube temperature was 80 ℃ and the flow rate of the gas was 2.0mL/min. Results:Under the above conditions, the linearity ranges of D-xylose, D-fructose, D-glu- cose, and sucrose were within 0.26-2.60mL/min, 0.28~2.80mL/min, 0. 14-1.40mL/min and 1.40-14.00 mL/min,respectively. The average recoveries were all within 95.0 % - 10.05 %, RSD were less than 2.0 %. Conclusion:This method is reliable, simple and reproducible, it is suitable for the determination of monosaccharide and sucrose in Astragalusinjection, which can provide reference of the quality control for Astragaius injection.
